Mouse Albumin ELISA kit ab108792 and Creatinine Assay Kit ab65340 used with mouse urine.

Szabo et al measured urine albumin and urine albumin creatinine ratio in wildtype, and Epcam and Tacstd2 null mice.

To collect urine, 2-3 week old mice were held over plastic sheet for up to 30 s with lightly stroking the belly to allow urination. Urine was then transferred into an Eppendorf tube, cleared by centrifugation, and stored at −80°C until analyzed. 1 µl of urine was used to measure albumin concentration by colorimetric ELISA assay (Mouse Albumin ELISA kit, ab108792, Abcam) according to the manufacturer's protocol. To determine creatinine concentration, 2 µl of urine were used in a colorimetric assay (Creatinine Assay Kit, ab65340, Abcam). At the conclusion of the assay, absorbances were read on Synergy Neo microplate reader (Agilent Biotek, Santa Clara, CA, USA). All standards and samples were measured in duplicates and the concentrations were determined from the average of the two values.

Urea Assay Kit ab83362 used to measure blood urea nitrogen with mouse serum, and Mouse Albumin ELISA kit ab108792 and Creatinine Assay Kit ab65340 used to measure albumin-creatinine ratio with mouse urine.

Widjaja et al subjected IL11 null mice and wild type controls to acute kidney injury and ran a blood urea nitrogen assay (Urea Assay Kit ab83362), and measured urine albumin-creatinine ratio using Mouse Albumin ELISA kit ab108792 and Creatinine Assay Kit ab65340.

Red indicates acute kidney injury. AKI was induced by intraperitoneal (IP) injection of folic acid (FA, 200 mg/kg) in vehicle (0.3 M NaHCO3) to 10–12-week-old male mice; control mice were administered vehicle alone.

All ELISA and colorimetric assays were performed according to the manufacturer’s protocol.

Mouse Albumin ELISA kit (ab108792) is a sandwich ELISA kit designed for the quantitative measurement of mouse Albumin levels in plasma, serum, urine, cell culture supernatant, cell lysate and tissue samples.

An Albumin specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently an Albumin specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Conjugate is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of Albumin captured in plate.

How the assay works

Albumin also known as ALB is a major plasma protein with a molecular mass of approximately 66.5 kDa. It predominantly gets synthesized in the liver and found abundantly in blood plasma. Albumin serves multiple functions including maintaining osmotic pressure and acting as a carrier protein for various endogenous substances like hormones fatty acids and bilirubin.
Biological function summary

Albumin plays a critical role in transporting small molecules like metabolites and drugs through the circulatory system. It is not a part of any complex but functions independently to bind transport and release various ligands. Albumin's affinity for hydrophobic molecules makes it essential for they to be solubilized in blood for proper metabolism and excretion.

Pathways

More than five binding sites on albumin facilitate its role in the fatty acid metabolism and renin-angiotensin system pathways. In fatty acid metabolism albumin interacts with transport proteins carrying fatty acids to tissues for energy production. Additionally during the renin-angiotensin system albumin modulates blood pressure through its interaction with angiotensin II which can influence vasoconstriction.

Researchers link albumin with nephrotic syndrome and liver cirrhosis. In nephrotic syndrome low levels of albumin (albumin low) can cause edema due to imbalanced osmotic pressure. In liver cirrhosis a reduction in albumin synthesis signifies liver damage. Albumin's interaction with proteins like angiotensin II correlates with complications such as hypertension further establishing the protein's role in health and disease contexts.

Major zinc transporter in plasma, typically binds about 80% of all plasma zinc (By similarity). Major calcium and magnesium transporter in plasma, binds approximately 45% of circulating calcium and magnesium in plasma (By similarity). Potentially has more than two calcium-binding sites and might additionally bind calcium in a non-specific manner (By similarity). The shared binding site between zinc and calcium at residue Asp-273 suggests a crosstalk between zinc and calcium transport in the blood (By similarity).
