Interpolated concentrations of native Alpha-synuclein in mouse serum, plasma (EDTA), plasma (heparin).
The concentrations of Alpha-synuclein were measured in duplicates, interpolated from the target standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 200, plasma (EDTA) 1 : 100, plasma (heparin) 1 : 200. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean target concentration was determined to be 101 ng/mL in serum, 57 ng/mL in plasma (EDTA), and 157 ng/mL in and plasma (heparin).

Undiluted samples are as follows : serum 12.5%, plasma (EDTA) 12.5%, plasma (citrate) 25%, and plasma (heparin) 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean target concentration was determined to be 4,055 pg/mL in serum, 3,930 pg/mL in plasma (EDTA), 2,132 pg/mL in plasma (citrate) and 4,658 pg/mL in and plasma (heparin).

How the assay works
Mouse Alpha-synuclein SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Alpha-synuclein is a 140- amino acid protein encoded by the SNCA gene, primarily found in neural tissue. It is a member of the synuclein family of proteins, including β-Synuclein and γ-Synuclein. Alpha-synuclein has been found concentrated in the presynaptic nerve terminals of neurons and has been found in the nucleus of neurons. Alpha-synuclein is a major component of Lewy Bodies. This protein appears to play a role in dopamine metabolism and vesicle trafficking.

Biological function summary
The alpha-synuclein protein plays critical roles in neuronal activity. It contributes to neurotransmitter release regulation by acting in the formation and plasticity of the presynaptic neuronal network. Alpha-synuclein doesn't usually form parts of large protein complexes but it may associate transiently with membranes and vesicular structures. The protein's monomer form has also been observed in alpha lines and related neuronal processes operating alongside various cellular functions.
Pathways
Synaptic vesicle trafficking and dopamine neurotransmitter release are significant areas involving the alpha-synuclein protein. In these pathways alpha-synuclein interacts with other proteins like synaptophysin and protein monomer monomerizations are intrinsic to these processes. Altered function or aggregation of alpha-synuclein disrupts these pathways influencing broader neurological functions.
