Mouse ALT ELISA Kit ab282882 is a rapid single-wash 90-min ELISA kit to measure Mouse ALT in serum and plasma. This SimpleStep ELISA® sensitivity is 4 pg/mL.
How the assay works
Mouse ALT SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Assay Specificity
This kit is designed for the quantification of mouse ALT. The standard protein in this kit is full length mouse ALT. Native signal was detected in serum, plasma (citrate), plasma (EDTA), and plasma (heparin)sample types. Cell culture supernatant, saliva, urine, milk, CSF, cell extract, and tissue extract samples have not been tested with this kit.

1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate for 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add stop solution

The ALT standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
Interpolated concentrations of native ALT in mouse serum, plasma samples.
The concentrations of ALT were measured in duplicates, interpolated from the ALT standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 2.5%, plasma – citrate 5%, plasma -heparin 5% and plasma – EDTA 5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ALT concentration was determined to be 67.60 ng/mL in serum, 30.36 ng/mL in plasma – citrate, 48.30 ng/mL in plasma – heparin and 38.86 ng/mL in plasma - EDTA.
Interpolated concentration of native ALT was measured in duplicate at different sample concentrations in 96-well vs. 384-well plates. Undiluted samples are 2.5% mouse serum. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer PTR.
