Interpolated concentrations of native Apolipoprotein E in mouse serum and plasma samples.

The concentrations of Apolipoprotein E were measured in duplicates interpolated from the Apolipoprotein E standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 56000 plasma (citrate) 1 : 56000 plasma (EDTA) 1 : 7000 and plasma (heparin) 1 : 56000. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Apolipoprotein E concentration was determined to be 155.61 µg/mL in neat serum 135.57 µg/mL in neat plasma (citrate) 132.0 µg/mL in neat plasma (EDTA) and 161.3 µg/mL in neat plasma (heparin).

How the assay works

Mouse Apolipoprotein E ELISA Kit, Chemiluminescent (ab326431) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

APOE is an apolipoprotein, a protein associating with lipid particles, that mainly functions in lipoprotein-mediated lipid transport between organs via the plasma and interstitial fluids. APOE is a core component of plasma lipoproteins and is involved in their production, conversion and clearance. Apolipoproteins are amphipathic molecules that interact both with lipids of the lipoprotein particle core and the aqueous environment of the plasma. As such, APOE associates with chylomicrons, chylomicron remnants, very low density lipoproteins (VLDL) and intermediate density lipoproteins (IDL) but shows a preferential binding to high-density lipoproteins (HDL). It also binds a wide range of cellular receptors including the LDL receptor/LDLR and the very low-density lipoprotein receptor/VLDLR that mediate the cellular uptake of the APOE-containing lipoprotein particles (By similarity). Finally, APOE also has a heparin-binding activity and binds heparan-sulfate proteoglycans on the surface of cells, a property that supports the capture and the receptor-mediated uptake of APOE-containing lipoproteins by cells (PubMed : 23676495).
