Mouse Biglycan ELISA kit (ab275906) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Mouse Biglycan protein in mouse serum, plasma and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse Biglycan with 193 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Biglycan is a proteoglycan found in the extracellular matrix and plays a key role in the maintenance of bone, cartilage, and tendons. Along with its structural role, biglycan plays a crucial role in inflammatory signaling during stress and tissue injury. It is currently under investigation as a potential target for clinical treatment of muscular dystrophies and certain inflammatory diseases such as osteoarthritis. The antibodies for this kit were raised against a recombinant protein representing the majority of the c-terminal portion of the mature protein. This protein is homologous throughout Mammalia, so cross-reactivities are likely for samples from many species.

Biglycan also known as BGN is a small leucine-rich proteoglycan with a molecular weight of roughly 38 kDa. Researchers frequently observe Biglycan in tissues such as bone cartilage and vasculature. The protein interacts with several other molecules in the extracellular matrix (ECM) including collagen and elastin to maintain the structural integrity of tissues. Through these interactions Biglycan plays an important role in supporting cell attachment and matrix organization.
The protein Biglycan influences cellular processes such as growth and differentiation. It is part of a complex network within the ECM that modulates cell signaling. Biglycan interacts with growth factors like transforming growth factor-beta (TGF-beta) and binds to Toll-like receptors (TLRs) affecting immune responses. These capabilities reveal Biglycan’s involvement in tissue development and repair.
Extracellular matrix modulation shows a significant role for Biglycan. It is involved in the TGF-beta signaling pathway and the inflammatory pathway mediated by TLRs. Through these pathways Biglycan interacts with proteins such as decorin and fibromodulin contributing to the regulation of collagen fibrillogenesis and inflammatory responses. Its role in these pathways highlights Biglycan’s influence in cellular communication and matrix regulation.
Biglycan has associations with osteoarthritis and muscular dystrophy. In osteoarthritis researchers observe altered expression levels of Biglycan which affects cartilage integrity. In muscular dystrophy the protein’s connection to dystrophin an important structural protein is noted. These associations illustrate Biglycan's impact on disease progression and its potential as a therapeutic target.

Interpolated concentrations of native Biglycan in mouse articular cartilage tissue extract based on a 75 μg/mL extract load.
The concentrations of Biglycan were measured in duplicate and interpolated from the Biglycan standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Biglycan concentration was determined to be 5,464.65 pg/mL.
