Sandwich ELISA - Mouse Calprotectin ELISA Kit (S100A8/S100A9) (AB263885)
Interpolated concentrations of native Calprotectin in mouse serum, plasma and tissue culture supernatant samples.
The concentrations of Calprotectin were measured in duplicates, interpolated from the Calprotectin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma (citrate) 50%, plasma (EDTA) 50%, plasma (heparin) 50% and lung supernatant 1 : 20. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Calprotectin concentration was determined to be 6,236 pg/mL in serum, 17,549 pg/mL in plasma (citrate), 52,502 pg/mL in plasma (EDTA), 3,225 pg/mL in plasma (Heparin), and 81,971 pg/mL in lung supernatant.

Product details
Mouse Calprotectin ELISA Kit (S100A8/S100A9) ab263885 is a rapid single-wash 90-min Sandwich ELISA to measure Mouse Calprotectin (S100A8/S100A9)in cell culture supernatant, citrate plasma, EDTA plasma, heparin plasma, serum. This SimpleStep sensitivity is 270.3 pg/mL.
How the assay works
Mouse Calprotectin (S100A8/S100A9)SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Assay Specificity
Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
Please refer to our protocol booklet for more details.

1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm
Calprotectin (S100A8/S100A9) is a heterodimer of the proteins S100A8 and S100A9. Calprotectin carries a wide variety of intra- and extracellular functions such as being involved in cytoskeletal structure, signal transduction, inflammation, antimicrobial activity, and apoptosis-inducing activities. Studies have shown calprotectin to have a prominent role in various pathophysiological processes in which targeting calprotectin may help prevent or minimize the severe effects of inflammation and associated diseases.

Biological function summary
The S100A8/S100A9 complex modulates inflammatory processes and immune responses. It acts as a pro-inflammatory mediator and is associated with leukocyte recruitment to sites of inflammation. This complex plays an important role in protecting cells from infections by inhibiting bacterial growth through sequestration of nutrient metals showcasing antimicrobial properties. Calprotectin significantly impacts immune responses and has become an interesting target for calprotectin ELISA kits to quantify its presence in plasma and other bodily fluids indicating inflammation.
Pathways
S100A8 and its partner S100A9 are involved in toll-like receptor and receptor for advanced glycation end-products (RAGE) signaling pathways. These pathways are key in mediating inflammatory responses and linking to innate immunity. The calgranulin A ELISA is often used to evaluate their involvement in these pathways helping to elucidate interactions with other proteins like calmodulin that play a role in cellular regulation processes.
