Interpolated concentrations of native Calprotectin in mouse serum, plasma and tissue culture supernatant samples.

The concentrations of Calprotectin were measured in duplicates, interpolated from the Calprotectin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma (citrate) 50%, plasma (EDTA) 50%, plasma (heparin) 50% and lung supernatant 1 : 20. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Calprotectin concentration was determined to be 6,236 pg/mL in serum, 17,549 pg/mL in plasma (citrate), 52,502 pg/mL in plasma (EDTA), 3,225 pg/mL in plasma (Heparin), and 81,971 pg/mL in lung supernatant.

Calprotectin (S100A8/S100A9) is a heterodimer of the proteins S100A8 and S100A9. Calprotectin carries a wide variety of intra- and extracellular functions such as being involved in cytoskeletal structure, signal transduction, inflammation, antimicrobial activity, and apoptosis-inducing activities. Studies have shown calprotectin to have a prominent role in various pathophysiological processes in which targeting calprotectin may help prevent or minimize the severe effects of inflammation and associated diseases.

S100A8 also known as calprotectin or MRP8 is a calcium-binding protein with a molecular weight of around 10.8 kDa. It is a member of the S100 protein family and is found expressed in myeloid cells such as neutrophils and monocytes. This protein often forms a heterodimer complex with S100A9 together referred to as calprotectin which plays a critical role in the immune response. Researchers frequently utilize calprotectin fecal ELISA MRP8 ELISA or calprotectin serum immunoassay to measure its levels in various biological samples.
Biological function summary

The S100A8/S100A9 complex modulates inflammatory processes and immune responses. It acts as a pro-inflammatory mediator and is associated with leukocyte recruitment to sites of inflammation. This complex plays an important role in protecting cells from infections by inhibiting bacterial growth through sequestration of nutrient metals showcasing antimicrobial properties. It can induce neutrophil chemotaxis, adhesion, can increase the bactericidal activity of neutrophils by promoting phagocytosis via activation of SYK, PI3K/AKT, and ERK1/2 and can induce degranulation of neutrophils by a MAPK-dependent mechanism (By similarity). Predominantly found as calprotectin (S100A8/A9) which has a wide plethora of intra- and extracellular functions (By similarity). The intracellular functions include : facilitating leukocyte arachidonic acid trafficking and metabolism, modulation of the tubulin-dependent cytoskeleton during migration of phagocytes and activation of the neutrophilic NADPH-oxidase (PubMed : 15331440). Participates also in regulatory T-cell differentiation together with CD69 (By similarity). Activates NADPH-oxidase by facilitating the enzyme complex assembly at the cell membrane, transferring arachidonic acid, an essential cofactor, to the enzyme complex and S100A8 contributes to the enzyme assembly by directly binding to NCF2/P67PHOX (By similarity).
