Sandwich ELISA - Mouse CCL21a ELISA Kit (AB208985)
Interpolated concentrations of native CCL21a in mouse serum and plasma samples.
The concentrations of CCL21a were measured in duplicates, interpolated from the CCL21a standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 2.5%, plasma (citrate) 2.5%, plasma (EDTA) 2.5%, and plasma (heparin) 2.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CCL21a concentration was determined to be 6.51 ng/mL in serum, 7.49 ng/mL in plasma (citrate), 5.97 ng/mL in plasma (EDTA) and 6.07 ng/mL in plasma (heparin).

Sandwich ELISA - Mouse CCL21a ELISA Kit (AB208985)
Interpolated concentrations of native CCL21a in mouse lung extract and mouse lung supernatant samples.
Interpolated concentrations of native CCL21a in mouse lung extract based on a 100 μg/mL extract load. The concentrations of CCL21a were measured in duplicates, interpolated from the CCL21a standard curves and corrected for sample dilution. Undiluted samples are as follows : mouse lung extract 100 μg/mL and mouse lung supernatant 6.25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CCL21a concentration was determined to be 0.61 ng/mL in mouse lung extract and 3.15 ng/mL in mouse lung supernatant. Mouse lung supernatant was cultured for 6 days in RPMI base media plus 10% fetal bovine serum (FBS).

Product details
Mouse CCL21a ELISA Kit (ab208985) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of CCL21a protein in cell culture extracts, cell culture supernatant, serum, tissue extracts, cit plasma, edta plasma, and hep plasma. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse CCL21a with 1 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Mouse CCL21a is a member of the CC subfamily of chemokines. Chemokines are a class of cytokines that induce chemotaxis of target cells. CCL21a is highly expressed in the lymph nodes and chemoattracts naïve T cells and mature dendritic cells into T zones of secondary lymphoid organs. Furthermore, it has been suggested that CCL21a induces human microvascular endothelial cell (HMVEC) migration and/or tube formation. In addition, it has been reported that sustained high concentrations of soluble or immobilized CCL21a inhibits T cell proliferation and IL-2 secretion. Human CCL21a exhibits 71% amino acid identity to mouse CCL21a.

Biological function summary
The actions of Ccl21a are important for the immune system's homeostasis and surveillance. It helps maintain the organized structure of lymphoid tissues by ensuring the correct positioning of lymphocytes. Ccl21a operates as part of the network that forms the microenvironment necessary for immune cell interactions and antigen presentation. There are no notable reports of it forming part of a larger protein complex but its receptor CCR7 is an essential player in its function.
Pathways
Ccl21a is involved in the chemokine signaling pathway and the regulation of immune cell trafficking. It interacts heavily within the pathway that involves the engagement of CCR7 a receptor that Ccl21a primarily activates alongside its counterpart ligand CCL19. Through these interactions Ccl21a influences processes related to lymphocyte migration and adaptive immune responses.
