Interpolated concentrations of native CD27 in mouse EL4-IL2 cell culture supernatant samples, both untreated and treated with 10 µg/ml PHA and 10 ng/ml PMA for two days.

The concentrations of CD27 were measured in duplicates, interpolated from the CD27 standard curves and corrected for sample dilution. Undiluted samples are as follows : untreated EL4-IL2 supernatant 100% and treated EL4-IL2 supernatant 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD27 concentration was determined to be 426 pg/mL in untreated EL4-IL2 supernatant and 1,978 pg/ml in treated EL4-IL2 supernatant.

The concentrations of CD27 were measured in duplicates, interpolated from the CD27 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 12.5%, plasma (citrate) 12.5%, plasma (heparin) 12.5%, and plasma (EDTA) 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD27 concentration was determined to be 14.3 ng/mL in serum, 11.9 ng/mL in plasma (citrate), 13.4 g/ml in plasma (heparin), and 10.8 ng/ml in plasma (EDTA).

Mouse CD27 ELISA Kit (ab233619) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of CD27 protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse CD27 with 10.9 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. CD27 also known as Tumor Necrosis Factor Receptor Superfamily Member 7 (TNFRSF7) is a protein chiefly involved in T cell activation and proliferation. It belongs to the TNF-receptor superfamily and weighs approximately 50 kDa. CD27 is expressed mainly on T lymphocytes and to a lesser extent on B cells and natural killer cells. Researchers often study CD27 and its ligand CD70 to understand immune responses. Tools such as anti-CD27 percp 7h21 monoclonal antibodies and APC-conjugated forms help detect this protein in various assays.
Biological function summary

One finds CD27 at the heart of cellular immune responses. It does not function alone; instead it typically forms part of a receptor-ligand complex with CD70 which influences T cell and B cell activities. Costimulatory immune-checkpoint receptor expressed at the surface of T-cells, NK-cells and B-cells which binds to and is activated by its ligand CD70/CD27L expressed by B-cells. The CD70-CD27 signaling pathway mediates antigen-specific T-cell activation and expansion which in turn provides immune surveillance of B-cells (PubMed : 11062504). Mechanistically, CD70 ligation activates the TRAF2-PTPN6 axis that subsequently inhibits LCK phosphorylation to promote phenotypic and transcriptional adaptations of T-cell memory (By similarity). In addition, activation by CD70 on early progenitor cells provides a negative feedback signal to leukocyte differentiation during immune activation and thus modulates hematopoiesis (PubMed : 15723067). Negatively regulates the function of Th2 lymphocytes in the adipose tissue (PubMed : 38386557).
