Interpolated concentrations of native Chromogranin A in mouse serum, plasma and cell culture supernatant samples.

The concentrations of Chromogranin A were measured in duplicates, interpolated from the Chromogranin A standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 2%, plasma (citrate) 2%, plasma (EDTA) 1.5%, plasma (heparin) 2% and pancreas tissue culture supernatant 12%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Chromogranin A concentration was determined to be 367 ng/mL in neat serum, 256 ng/mL in neat plasma (citrate), 443 ng/mL in neat plasma (EDTA), 203 ng/mL in neat plasma (heparin) and 53 ng/mL in neat pancreas tissue culture supernatant.

Mouse Chromogranin A ELISA Kit (ab264631) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Chromogranin A protein in cell culture media, cell culture supernatant, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse Chromogranin A with 93 pg/ml sensitivity.

This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Chromogranin A (CHGA) a secreted acidic protein and a member of the chromogranin/secretogranin family of neuroendocrine secretory proteins. It is located in secretory vesicles of neurons and endocrine cells. It is processed to number of non-overlapping peptides including Beta-Granin, Pancreastatin, Catestatin and various forms of Serpins. Chromogranin A is a useful serological and immunohistological marker for the presence of neuroendocrine tumors.

Chromogranin A (CgA) sometimes called chromagranin A is a glycoprotein with a molecular weight of approximately 49–52 kDa. It is highly expressed in the secretory granules of neuroendocrine cells present in tissues such as the adrenal medulla pancreas and endocrine cells of the gastrointestinal tract. CgA identified as part of a family including chromogranin B and chromogranin C functions mechanically as a precursor to various bioactive peptides. This translates into its involvement in the storage and release of hormones and peptides within vesicles.
Biological function summary

Chromogranin A assists in the regulation of secretory pathways across multiple neuroendocrine systems. It plays a role in hormone storage conversion to active peptides and regulates the exocytosis of hormones. Inhibits catecholamine release from chromaffin cells and noradrenergic neurons by acting as a non-competitive nicotinic cholinergic antagonist. Can induce mast cell migration, degranulation and production of cytokines and chemokines.. Serpinin. Regulates granule biogenesis in endocrine cells by up-regulating the transcription of protease nexin 1 (SERPINE2) via a cAMP-PKA-SP1 pathway. This leads to inhibition of granule protein degradation in the Golgi complex which in turn promotes granule formation (PubMed : 21436258). Pyroglutaminated (pGlu)-serpinin exerts an antiapoptotic effect on cells exposed to oxidative stress (PubMed : 21537909).
