Sandwich ELISA - Mouse COX2 ELISA Kit (AB210574)
Interpolated concentrations of native mouse COX2 in an unstimulated and LPS (1 µg/mL, 6 hr) stimulated Raw 264.7 cell extract based on a 100 µg/mL extract load.
The concentrations of mouse COX2 were measured in duplicate and interpolated from the COX2 standard curve then corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean COX2 concentration was determined to be 0.204 ng/mL in unstimulated extract and 8.7 ng/mL in LPS stimulated Raw 264.7 extract.

Sandwich ELISA - Mouse COX2 ELISA Kit (AB210574)
Interpolated concentrations of native mouse COX2 in an unstimulated and LPS (1 µg/mL, 6 hr) stimulated Raw 264.7 cell extract based on a 100 and 50 μg/mL extract load, respectively.
The concentrations of mouse COX2 were measured in duplicate and interpolated from the COX2 standard curve then corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean COX2 concentration was determined to be 0.203 ng/mL in unstimulated extract and 5.3 ng/mL in LPS stimulated Raw 264.7 extract.

Product details
Mouse COX2 ELISA Kit (ab210574) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of COX2 protein in cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse COX2 with 97 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Cyclooxygenase-2 (COX2) also known as prostaglandin G/H synthase 2 (PGHS2) is a 70 kDa microsomal enzyme that belongs to the prostaglandin G/H synthase family. It is inducibly-expressed by a number of cell types, including fibroblasts, vascular smooth muscle cells, endothelium, and monocytes. Functionally, COX2 is a homodimer that catalyzes two steps in the conversion of arachadonic acid to prostaglandin H2. Mature mouse COX2 is 587 amino acids (aa) in length and contains one EGF-like domain (aa 18‑55).

Biological function summary
COX2 plays a significant role in the inflammatory response and is part of the complex process of synthesizing prostaglandins. These compounds mediate inflammation and pain making COX2 an important target for understanding these processes. COX2 is not ubiquitously expressed but rather is induced in activated macrophages and other cells during inflammatory conditions. Its function is also important for normal physiological processes like ovulation and implantation.
Pathways
COX2 is essential in the prostaglandin biosynthesis pathway connecting it to the arachidonic acid metabolism pathway. Cyclooxygenase 2 works with phospholipase A2 which releases arachidonic acid from the phospholipid membrane. COX2 then converts this acid to prostaglandin H2 a precursor for other prostaglandins. COX1 the other isoform of cyclooxygenase is closely related to COX2 and while they have different expression patterns they share some functional similarities in these pathways.
