The Mouse CXCL1 Enzyme-Linked Immunosorbent Assay (ELISA) kit (GRO alpha) (ab213859) is designed for the quantitative detection of Mouse CXCL1 (GRO alpha) in cell culture supernatants and serum.

The ELISA kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for CXCL1 (GRO alpha) has been precoated onto 96-well plates. Standards (Expression system for standard: E.coli; Immunogen sequence: R20-K96) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for CXCL1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Mouse CXCL1 amount of sample captured in plate.

Chemokine (C-X-C motif) ligand 1 (CXCL1) is a small cytokine belonging to the CXC chemokine family that was previously called GRO1 oncogene, GRO alpha, KC, Neutrophil-activating protein 3 (NAP-3) and melanoma growth stimulating activity, alpha (MSGA-alpha). In humans, this protein is encoded by the CXCL1 gene. The gene for CXCL1 is located on human chromosome 4 amongst genes for other CXC chemokines. The mature form of CXCL1 is maximally 73 amino acids long. CXCL1 is secreted by human melanoma cells, has mitogenic properties and is implicated in melanoma pathogenesis. CXCL1 is expressed by macrophages, neutrophils and epithelial cells, and has neutrophil chemoattractant activity. This chemokine elicits its effects by signaling through the chemokine receptor CXCR2. CXCL1 decreased the severity of multiple sclerosis and may offer a neuroprotective function.

Activity summary

CXCL1 also known as GRO alpha or KC in certain species is a chemokine with a molecular mass of around 7.8 kDa. This protein is primarily secreted by macrophages neutrophils and endothelial cells. It plays a critical role in the recruitment of neutrophils during inflammation and acts by binding to the CXCR2 receptor. CXCL1 is expressed in many tissues including lungs liver and skin where it regulates the immune response and facilitates tissue repair.

Biological function summary

CXCL1 functions as a significant mediator in inflammatory processes. It does not form a complex but acts independently to exert its effects. By attracting neutrophils to sites of injury or infection CXCL1 supports the body's defense mechanisms. This chemokine also influences angiogenesis contributing to the formation of new blood vessels which is essential in wound healing and tissue regeneration.

Pathways

CXCL1 plays a role in both the NF-kB signaling and MAPK signaling pathways. These pathways are pivotal for the regulation of immune and inflammatory responses. CXCL1 acts alongside proteins such as IL-8 another chemokine that also binds to the CXCR2 receptor. Through these pathways CXCL1 influences the activation and migration of immune cells facilitating a rapid response to inflammatory stimuli.

Associated diseases and disorders

CXCL1 has a significant association with conditions such as chronic obstructive pulmonary disease (COPD) and rheumatoid arthritis. In COPD elevated levels of CXCL1 contribute to persistent inflammation and neutrophil infiltration. In rheumatoid arthritis CXCL1 participates in joint inflammation and damage interacting with proteins like TNF-alpha which amplifies inflammatory responses. Understanding the role of CXCL1 in these conditions could help guide the development of targeted therapies to mitigate inflammation and tissue damage.
