Interpolated concentration of native Cytokeratin 18 was measured in duplicate at different sample concentrations. Undiluted samples are as follows : serum 50% and plasma (heparin) 50%. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.

Interpolated concentrations of native Cytokeratin 18 in mouse salivary gland and liver tissue extract based on a 5 µg/mL and 50 µg/mL extract load respectively.

The concentrations of Cytokeratin 18 were measured in duplicate and interpolated from the Cytokeratin 18 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Cytokeratin 18 concentration was determined to be 12131 pg/mL in salivary gland and 1404 pg/mL in liver tissue extracts.

How the assay works

Mouse Cytokeratin 18 ELISA Kit, Chemiluminescent (ab326400) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Target data

Required for the formation of KRT8/KRT18 filaments that are involved in ARHGEF40-mediated actin stress fiber formation and tensional force-induced stress fiber formation and reinforcement (By similarity). Also acts downstream of ROCK kinase activation as part of a positive feedback mechanism in response to cellular mechanical stress loading (By similarity). Organization and orientation of KRT18 filaments are responsible for the properly elongated morphology of epithelial tubules (By similarity). Involved in the uptake of thrombin-antithrombin complexes by hepatic cells (By similarity). When phosphorylated, plays a role in filament reorganization. Involved in the delivery of mutated CFTR to the plasma membrane. Together with KRT8, is involved in interleukin-6 (IL-6)-mediated barrier protection (PubMed : 17213200).
