Abcam's DPPIV (CD26) Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay designed for the quantitative measurement of mouse DPPIV (CD26) in serum, plasma and cell culture supernatants.

This assay employs an antibody specific for DPPIV coated on a 96-well plate. Standards and samples are pipetted into the wells and DPPIV present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse DPPIV antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of DPPIV bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

The enzyme Dipeptidyl Peptidase IV (DPPIV) also known as CD26 is a serine protease with an approximate mass of 110 kDa. It is expressed in various tissues throughout the body including the kidney liver and small intestine. DPPIV plays a significant role in cleaving dipeptides from the N-terminus of polypeptides which influences the activity of many bioactive peptides and proteins.
Biological function summary

DPPIV functions in the regulation of immune responses and glucose metabolism. It is not typically part of a complex structure operating independently to modulate peptide hormones and cytokines. Cell surface glycoprotein receptor involved in the costimulatory signal essential for T-cell receptor (TCR)-mediated T-cell activation. Acts as a positive regulator of T-cell coactivation, by binding at least ADA, CAV1, IGF2R, and PTPRC. Its binding to CAV1 and CARD11 induces T-cell proliferation and NF-kappa-B activation in a T-cell receptor/CD3-dependent manner. Its interaction with ADA also regulates lymphocyte-epithelial cell adhesion. In association with FAP is involved in the pericellular proteolysis of the extracellular matrix (ECM), the migration and invasion of endothelial cells into the ECM. May be involved in the promotion of lymphatic endothelial cells adhesion, migration and tube formation. When overexpressed, enhanced cell proliferation, a process inhibited by GPC3. Also acts as a serine exopeptidase with a dipeptidyl peptidase activity that regulates various physiological processes by cleaving peptides in the circulation, including many chemokines, mitogenic growth factors, neuropeptides and peptide hormones. Removes N-terminal dipeptides sequentially from polypeptides having unsubstituted N-termini provided that the penultimate residue is proline.
