Sandwich ELISA - Mouse Epcam ELISA Kit (AB242247)
Interpolated concentrations of native Epcam in mouse F9 cell extract and 4T1 cell extract based on an 80 μg/mL and 35 μg/mL extract load, respectively. The concentrations of Epcam were measured in duplicate and interpolated from the Epcam standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Epcam concentration was determined to be 5,449 pg/mL in F9 cells and 6774 pg/mL in 4T1 cells.

Product details
Mouse Epcam ELISA Kit (ab242247) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Epcam protein in cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse Epcam with 6 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Epcam, or epithelial cell adhesion molecule, is a membrane barrier glycoprotein that serves as a key modulator of cell to cell adhesion. When cleaved, this protein forms an extracellular and intracellular fragment, both of which have activate signaling cascades involved in cell growth and motility. This protein is currently under investigation for its tumorigenic effects and has been noted to be up-regulated in many epithelial cancers.

Biological function summary
EpCAM facilitates signaling pathways that impact cellular processes like proliferation and differentiation. It is not a part of a larger protein complex but interacts directly with other cellular machinery to transmit intracellular signals. EpCAM influences stem cell behavior and is important in tissue homeostasis assisting in both normal regenerative processes and wound healing.
Pathways
EpCAM participates in regulatory mechanisms involving the Wnt/β-catenin signaling pathway which affects gene expression and cell fate decisions. Through its action EpCAM modulates components like E-cadherin which is important for cell adhesion and the maintenance of the epithelial layer. It indirectly influences pathways relating to MYC a transcription factor that regulates cell cycle progression and apoptosis.
