MouseFGL2/Prothrombinase ELISA Kit (ab283884) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of FGL2/Prothrombinase protein in serum, plasma, and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse FGL2/Prothrombinase with 0.03 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. FGL2 (Fibrinogen-Like Protein 2, also known as Fibroleukin and Prothrombinase exists as both membrane bound and secreted by cytotoxic and helper T-cells (CD8+ and CD4+). By converting prothrombin to thrombin, it contributes to physiologic lymphocyte functions at mucosal sites.
FGL2 also known as Fibroleukin or Prothrombinase is a protein involved in various physiological and pathological processes. Its molecular mass is approximately 70 kDa. This protein is widely expressed in different tissues including the liver and the spleen as well as in immune cells such as T cells and macrophages. It serves as an important signaling molecule and mediator in various biological pathways due to its capacity to interact with other proteins and molecular complexes.
FGL2 acts in immune response modulation and blood coagulation. It can exist as a homodimer or form part of a larger protein complex. One of its roles involves cleaving prothrombin to generate thrombin contributing to the coagulation cascade. Moreover FGL2 has immunosuppressive properties affecting T cell proliferation and cytokine release. Its dual function in coagulation and immune regulation points to its importance in maintaining homeostasis and orchestrating immune responses.
FGL2 serves key roles in both the coagulation and immune pathways. In the coagulation pathway FGL2 aids in the conversion of prothrombin to thrombin bridging innate immunity with coagulation processes. This action places it in close functional relationship with proteins in the coagulation pathway such as thrombin and fibrin. Moreover its involvement in immune pathways aligns it with cytokines and interferons modulating the immune response and maintaining immune system balance.
FGL2 plays significant roles in conditions such as viral hepatitis and certain autoimmune disorders. In viral hepatitis FGL2's upregulation contributes to liver fibrosis and inflammation often in conjunction with another protein CD147. In autoimmune disorders the dysregulation of FGL2 can lead to excessive inflammatory responses impacting proteins like the interferon-gamma. Understanding FGL2's involvement in these diseases can inform therapeutic strategies targeting both coagulation and immune dysregulation.

The FGL2/Prothrombinase standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
Interpolated concentrations of native FGL2/Prothrombinase in mouse serum, plasma (EDTA), plasma (citrate), and plasma (heparin).
The concentrations of FGL2/Prothrombinase were measured in duplicates, interpolated from the target standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 25%, plasma (EDTA) 50%, plasma (citrate) 6.3%, and plasma (heparin) 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean target concentration was determined to be 37.3 ng/mL in neat serum, 20.3 ng/mL in neat plasma (EDTA), 45.8 ng/mL in neat plasma (citrate), and 34.0 ng/mL in plasma (heparin).
