Interpolated concentration of native Fibrinogen was measured in duplicate at different sample concentrations. Undiluted samples are as follows : serum 5%, lung tissue culture supernatant 5%, and urine 6.25%. The interpolated dilution factor corrected values to neat sample are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.

Percent reactivity against related fibrinogen proteins.

The following recombinant/native proteins were prepared at 50 ng/mL and assayed for cross-reactivity. Percent reactivity is determined by interpolating the protein concentrations from the standard curve and expressing the interpolated concentrations as a percentage of the protein concentration assayed at the same concentration.

Mouse Fibrinogen SimpleStep ELISA® kit is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Fibrinogen protein in Serum, Cit Plasma, EDTA Plasma, Hep Plasma, Cell culture supernatant, Urine. Quantitate Mouse Fibrinogen with 102 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

CROSS REACTIVITY

The following recombinant proteins were prepared at 50 ng/mL and 8,000 pg/mL and assayed for cross reactivity and no cross reactivity was observed: Human Fibrinogen alpha, Human Fibrinogen beta, Human Fibrinogen gamma, Human Factor XII, Human Plasmin, Human Thrombin, Rat Fibrinogen

The following recombinant proteins were prepared at 50 ng/mL and 8,000 pg/mL and assayed for cross reactivity.

Human D-Dimer: 27% cross-reactivity

SPECIES REACTIVITY

This kit recognizes human and mouse Fibrinogen protein.

Native purified human Fibrinogen was prepared at 18 ng/mL and assayed for cross reactivity. 145% cross reactivity was observed.

Other species reactivity was determined by measuring plasma – EDTA samples of various species, interpolating the protein concentrations from the mouse standard curve, and expressing the interpolated concentrations as a percentage of the protein concentration in mouse plasma assayed at the same dilution.

Reactivity was determined for the following species: Human (147%), Monkey (9%)

No signal was observed for the following species: Rat, Cow

Cleaved by the protease thrombin to yield monomers which, together with fibrinogen beta (FGB) and fibrinogen gamma (FGG), polymerize to form an insoluble fibrin matrix. Fibrin has a major function in hemostasis as one of the primary components of blood clots (PubMed : 7649481). In addition, functions during the early stages of wound repair to stabilize the lesion and guide cell migration during re-epithelialization (PubMed : 11389004). Was originally thought to be essential for platelet aggregation, based on in vitro studies using anticoagulated blood (PubMed : 7649481). However, subsequent studies have shown that it is not absolutely required for thrombus formation in vivo (PubMed : 10930441). Enhances expression of SELP in activated platelets via an ITGB3-dependent pathway (PubMed : 19332769). Maternal fibrinogen is essential for successful pregnancy (PubMed : 7649481). Fibrin deposition is also associated with infection, where it protects against IFNG-mediated hemorrhage (PubMed : 12629066). May also facilitate the immune response via both innate and T-cell mediated pathways (PubMed : 23487423).
