IFN-induced antiviral protein which acts as an inhibitor of cellular as well as viral processes, cell migration, proliferation, signaling, and viral replication. Enhances MAVS-mediated host antiviral responses by serving as an adapter bridging TBK1 to MAVS which leads to the activation of TBK1 and phosphorylation of IRF3 and phosphorylated IRF3 translocates into nucleus to promote antiviral gene transcription. Exhibits an antiproliferative activity via the up-regulation of cell cycle negative regulators CDKN1A/p21 and CDKN1B/p27. Normally, CDKN1B/p27 turnover is regulated by COPS5, which binds CDKN1B/p27 in the nucleus and exports it to the cytoplasm for ubiquitin-dependent degradation. IFIT3 sequesters COPS5 in the cytoplasm, thereby increasing nuclear CDKN1B/p27 protein levels. Up-regulates CDKN1A/p21 by down-regulating MYC, a repressor of CDKN1A/p21. Can negatively regulate the apoptotic effects of IFIT2 (By similarity).

Mouse Flt-3 ELISA Kit (ab256395) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Flt-3 protein in cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse Flt-3 with 0.915 ng/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Flt-3, also known as CD135, is a receptor type tyrosine kinase involved in the differentiation of hematopoietic cells. Binding of its ligand, FLT3L induces phosphorylation cascades through multiple kinases, including STAT5A and MAPK1. Flt-3 is used to differentiate multipotent progenitor cells and hematopoietic stem cells. Mutations in this gene are linked to acute myelogenous leukemia and its levels are negatively associated with survival. Given Flt-3’s importance in this disease, it is currently being investigated as a therapeutic target.

IFIT3 also known as P60 is a member of the IFIT (Interferon-Induced Proteins with Tetratricopeptide Repeats) family of proteins. With a molecular mass of approximately 60 kDa IFIT3 is part of the antiviral response system. This protein is expressed in various tissues and is highly inducible by type I interferons. It functions to inhibit viral replication by interacting with viral RNA. The structure of IFIT3 includes tetratricopeptide repeat motifs which facilitate its role in protein-protein interactions.
IFIT3 plays a major role in the immune response against viral infections. The protein forms complexes with other IFIT family members like IFIT1 and IFIT2 to enhance antiviral efficacy. IFIT3 binds to foreign RNA lacking proper cap structures blocking viral protein synthesis. The protein's actions help strengthen the innate immune system and further control the spread of viral pathogens. IFIT3 also modulates cellular functions by engaging with signaling pathways that regulate inflammation and immune defense.
IFIT3 integrates into important signaling networks such as the interferon-mediated signal transduction pathway and the JAK-STAT signaling pathway. In these pathways IFIT3 along with related proteins like IRF3 and ISG15 helps amplify immune responses after detecting viral threats. Its involvement in these pathways ensures that antiviral states within cells are quickly established reducing viral survival and dissemination.
IFIT3 has significant implications for infection and cancer outcomes. In viral infections such as hepatitis C IFIT3 expression levels correlate with effective antiviral activity and prognosis. In cancer aberrant IFIT3 expression can influence tumorigenesis and impact treatment responses. Collaborating with proteins such as STAT1 and p53 IFIT3 can affect pathways involved in cell proliferation and apoptosis linking immune responses to cancer progression and therapy resistance.

The Flt-3 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
Interpolated concentrations of native Flt-3 in mouse M1 cell extract based on a 200 μg/mL extract load.
The concentrations of Flt-3 were measured in duplicate and interpolated from the Flt-3 standard curve and corrected for sample dilution. The interpolated, dilution factor corrected, values are plotted (mean +/- SD, n=2). The mean Flt-3 concentration was determined to be 40.5 ng/mL.
