Interpolated concentrations of native Flt3 ligand in mouse serum and plasma samples.
The concentrations of Flt3 ligand were measured in duplicates, interpolated from the Flt3 ligand standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 100%, plasma (citrate) 100%, plasma (EDTA) 100%, plasma (heparin) 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Flt3 ligand concentration was determined to be 232.95 pg/mL in serum, 163.28 pg/mL in plasma (citrate), 184.73 pg/mL in plasma (EDTA)and 269.77 pg/mL in plasma (heparin).

Mouse FLT3 Ligand ELISA kit (ab275551) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Mouse FLT3 Ligand protein in serum, plasma, and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse FLT3 Ligand with 3.9 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Flt-3 ligand is an alpha-helical cytokine that promotes the differentiation of multiple hematopoietic cell lineages. Mature human Flt-3 ligand consists of a 158 amino acid extracellular domain with a cytokine-like domain and a juxtamembrane tether region, a 21 aa transmembrane segment, and a 30 aa cytoplasmic tail. Flt-3 ligand is expressed as a noncovalently-linked dimer by T cells and bone marrow and thymic fibroblasts. Both transmembrane and soluble Flt-3 ligand signal through the tyrosine kinase receptor Flt-3/Flk-2. Flt-3 ligand induces the expansion of monocytes and immature dendritic cells as well as early B cell lineage differentiation. It synergizes with IL-3, GM-CSF, and SCF to promote the mobilization and myeloid differentiation of hematopoietic stem cells. Animal studies report Flt-3 ligand to reduce the severity of experimentally induced allergic inflammation. Human Flt-3 ligand shares 71% and 65% aa sequence identity with mouse and rat Flt-3 Ligand, respectively.

Biological function summary
Flt3 functions as a receptor that is activated by binding to its ligand FLT3 ligand (FLT3L). This interaction stimulates the associated tyrosine kinase activity leading to auto-phosphorylation of Flt3 and subsequent downstream signaling. Flt3 does not form a heterocomplex rather it dimerizes upon ligand binding to initiate signaling cascades. These cascades promote cell proliferation differentiation and survival particularly impacting hematopoiesis and immune responses.
Pathways
Flt3 signaling impacts the internal signaling networks involving the MAPK/ERK and PI3K/AKT pathways both central to cell cycle regulation and apoptosis. Flt3 interacts functionally with proteins such as SHP2 and GRB2 in these pathways aiding signal transduction. The Flt3 receptor also shares functional motifs with KIT and PDGF receptors indicating shared regulatory mechanisms involved in cellular proliferation.
