Interpolated concentrations of native Granzyme A in mouse splenocytes culture supernatant samples.
The concentrations of Granzyme A were measured in duplicates, interpolated from the Granzyme A standard curves and corrected for sample dilution. Undiluted samples are as follows : spleen supernatant 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Granzyme A concentration was determined to be 615 pg/mL in neat splenocyte culture supernatant.

Mouse Granzyme A ELISA Kit (ab253213) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Granzyme A protein in cell culture supernatant, cit plasma, edta plasma, serum, and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse Granzyme A with 11 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Granzyme A, also known as T cell-specific serine protease 1, is a trypsin-like serine protease that forms a disulfide-linked homodimer and is secreted by cytotoxic T lymphocytes and natural killer cells. While Granzyme A is the most abundant serine protease in killer cell granules, it is considered less cytotoxic than Granzyme B, but can activate the caspase-independent cell death with morphological features of apoptosis. Granzyme A targets several nuclear proteins for degradation including, Pro-IL-1beta, NDUFS3, SET, AP1, Histone H1, HMGB2, PARP-1, Ku70, and lamins, thus limiting DNA repair, oxidative repair activity, and disrupting the election transport of mitochondria. Granzyme A is also considered to have extracellular functions including promoting inflammation and degradation of extracellular matrix.

Biological function summary
Functions of Granzyme A extend to inducing apoptosis in target cells alongside its role in promoting inflammation. It operates independent of caspases and directly cleaves substrates in the cell to induce cell death making it different from the related Granzyme B. As part of the granzyme-perforin complex it enters target cells via perforin-created pores mediates reactions leading to substrate degradation and promotes DNA damage.
Pathways
Granzyme A gets involved in the granule exocytosis pathway which constitutes an important mechanism for CTLs and NK cells to kill infected or cancerous cells. The protein is also linked to the inflammatory response pathways where it can interact with other serine proteases. Related proteins like perforin and Granzyme B also engage in these pathways and they collectively work to trigger apoptosis and modulate immune responses.
