Mouse GREM2 ELISA Kit is designed for the quantitative determination of GREM2 in cell culture supernatants, plasma and serum samples.

This assay employs an antibody specific for mouse GREM2 coated on a 96-well plate. Standards and samples are pipetted into the wells and GREM2 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse GREM2 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of GREM2 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Activity summary

PRDC also known as protein related to DAN and cerberus or GREM2 is a member of the DAN family of cysteine knot secreted proteins. It weighs approximately 24 kDa. PRDC expresses in various tissues with strong presence in the heart lung and kidney among others. Its expression pattern suggests a role in organ development and tissue homeostasis. As a secreted protein PRDC exists outside the cell where it can interact with other molecules in the extracellular environment.

Biological function summary

PRDC functions as an antagonist of bone morphogenetic proteins (BMPs) which are part of the TGF-beta superfamily. By inhibiting BMP signaling PRDC regulates cell differentiation and growth. This protein does not work alone; it often interacts within larger complexes that involve other members of the BMP family. Through this interaction PRDC plays a role in developmental processes and tissue regeneration.

Pathways

PRDC integrates into key signaling cascades such as the BMP signaling pathway and TGF-beta pathway. It modulates the activity of BMPs directly affecting SMAD-dependent signaling. This role places it as a critical player in embryogenesis and organogenesis. Proteins like noggin and chordin also intersect with PRDC within these pathways working together to modulate cellular responses.

Associated diseases and disorders

Altered PRDC function links to various conditions including fibrodysplasia ossificans progressiva (FOP) and pulmonary hypertension. These diseases highlight the importance of proper BMP signaling modulation by PRDC. In the context of FOP abnormal PRDC expression can exacerbate processes like heterotopic ossification. Proteins such as ACVR1 involved in BMP signaling exhibit interconnected roles with PRDC in these pathological conditions.
