Abcam's HGF Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of mouse HGF in cell lysates and tissue lysates.
This assay employs an antibody specific for mouse HGF coated on a 96- well plate. Standards and samples are pipetted into the wells and HGF present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse HGF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of HGF bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Biological function summary
HGF plays an essential role in cellular growth regeneration and healing processes across different tissues. It is not part of a complex but functions as a single-chain polypeptide that undergoes cleavage into its active form. HGF facilitates cell proliferation and migration; this is especially important during organ regeneration and repair processes. By engaging with its receptor c-Met it induces cell survival and cytoskeletal reorganization making it important for wound healing.
Pathways
HGF is critical in the MAPK and PI3K-Akt signaling pathways which impact processes like cell division and survival. Through these pathways HGF interacts with other proteins such as ERK and Akt contributing to the regulation of cellular growth and metabolism. HGF's role in these pathways highlights its importance in developmental processes and tissue homeostasis.
