Interpolated concentrations of native HIF-1 alpha in mouse deferoxamine treated versus untreated NIH/3T3 cell extract based on a 10 µg/mL extract load. The concentrations of HIF1a were measured in duplicate, interpolated from the HIF1a standard curves and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean HIF1a concentration was determined to be 485.55 pg/mL and 40.77 pg/ml in deferoxamine and mock treated cell extract, respectively.

How the assay works

Mouse HIF-1 alpha ELISA Kit, Chemiluminescent (ab325897) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Target data

Functions as a master transcriptional regulator of the adaptive response to hypoxia (PubMed : 15225651, PubMed : 17981124, PubMed : 22009797). Under hypoxic conditions, activates the transcription of over 40 genes, including erythropoietin, glucose transporters, glycolytic enzymes, vascular endothelial growth factor, HILPDA, and other genes whose protein products increase oxygen delivery or facilitate metabolic adaptation to hypoxia (PubMed : 15225651, PubMed : 17981124, PubMed : 22009797, PubMed : 22100406, PubMed : 37558878). Plays an essential role in embryonic vascularization, tumor angiogenesis and pathophysiology of ischemic disease (PubMed : 22009797). Heterodimerizes with ARNT; heterodimer binds to core DNA sequence 5'-TACGTG-3' within the hypoxia response element (HRE) of target gene promoters (PubMed : 26245371). Activation requires recruitment of transcriptional coactivators such as CREBBP and EP300. Activity is enhanced by interaction with NCOA1 and/or NCOA2. Interaction with redox regulatory protein APEX1 seems to activate CTAD and potentiates activation by NCOA1 and CREBBP. Involved in the axonal distribution and transport of mitochondria in neurons during hypoxia (By similarity).
