Product details
The Mouse IL1RL1 Enzyme-Linked Immunosorbent Assay (ELISA) kit (ab213871) is designed for the quantitative measurement of Mouse IL1RL1 in cell culture supernatants, cell lysates, tissue homogenates, serum and plasma (heparin, EDTA).
The ELISA kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for IL1RL1 has been pre-coated onto 96-well plates. Standards (Expression system for standard: NSO; Immunogen sequence: S27-R332) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for IL1RL1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Mouse IL1RL1 amount of sample captured in plate.
Interleukin 1 receptor-like 1, also known as IL1RL1 or ST2, is a protein that in humans is encoded by the IL1RL1 gene. The protein encoded by this gene is a member of the interleukin 1 receptor family. Studies of the similar gene in mouse suggested that this receptor can be induced by proinflammatory stimuli, and may be involved in the function of helper T cells. IL1RL1 is necessary for endotoxin tolerance and, by inhibiting TLR responses, enhances Th2 responses. This gene, interleukin 1 receptor, type I (IL1R1), interleukin 1 receptor, type II (IL1R2) and interleukin 1 receptor-like 2 (IL1RL2) form a cytokine receptor gene cluster in a region mapped to chromosome 2q12.

Biological function summary
ST2 interacts with interleukin-33 (IL-33) and functions as its receptor. The binding of IL-33 to ST2 activates immune responses particularly influencing T-helper 2 (Th2) cells. ST2 forms part of a receptor complex including IL-1 receptor accessory protein IL-1RAcP necessary for signal transduction. This interaction leads to the activation of nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kB) and mitogen-activated protein kinase (MAPK) signaling pathways facilitating inflammatory responses.
Pathways
IL-33 signals through the ST2 receptor participating actively in the immune signaling pathway. This pathway plays a significant role in modulating inflammatory and immune responses. ST2 association with NF-kB broadly affects inflammatory signaling while its involvement with the MAPK pathway aids in cellular responses to cytokines and stress. The close relationship with the IL-33/ST2 axis highlights its importance in maintaining immune homeostasis and reacting to environmental stressors.
