Linearity of dilution is determined based on interpolated values from the standard curve. Linearity of dilution defines a sample concentration interval in which interpolated target concentrations are directly proportional to sample dilution.

Recombinant IL-18 was spiked into the following biological samples and diluted in a 2-fold dilution series in Sample Diluent NS.

Interpolated concentrations of native IL-18 in mouse serum and plasma samples.

The concentrations of IL-18 were measured in duplicate and interpolated from the IL-18 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean IL-18 concentration was determined to be 128 pg/mL serum 135pg/mL in plasma (citrate) 101pg/mL in plasma (EDTA) and 190pg/mL in plasma (heparin).

How the assay works

Mouse IL-18 ELISA Kit, Chemiluminescent - Extracellular (ab327073) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Target data

Pro-inflammatory cytokine primarily involved in epithelial barrier repair, polarized T-helper 1 (Th1) cell and natural killer (NK) cell immune responses (PubMed : 26638072, PubMed : 26638073). Upon binding to IL18R1 and IL18RAP, forms a signaling ternary complex which activates NF-kappa-B, triggering synthesis of inflammatory mediators (By similarity). Synergizes with IL12/interleukin-12 to induce IFNG synthesis from T-helper 1 (Th1) cells and natural killer (NK) cells (By similarity). Involved in transduction of inflammation downstream of pyroptosis : its mature form is specifically released in the extracellular milieu by passing through the gasdermin-D (GSDMD) pore (PubMed : 30392956).
