Interpolated concentrations of native iNOS in RAW264.7 cell extract samples based on a 400 μg/mL extract load.
The concentrations of iNOS were measured in duplicate and interpolated from the iNOS standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean iNOS concentration was determined to be 1,052.8 pg/mL in RAW264.7 cell extract samples.

Mouse iNOS ELISA Kit (ab253219) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of iNOS protein in cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse iNOS with 18.1 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

iNOS, also known as NOS2, catalyzes the production of nitric oxide from L-arginine, which is used in various cell signaling events. iNOS is an inducible member of the NOS family and is upregulated as a host-defense mechanism during proinflammatory cytokine activity seen with tumorigenesis and parasitic infection. iNOS activity is a key cause of septic shock.

Biological function summary
INOS plays a significant role in the immune system by participating in the defense against pathogens. The enzyme is part of a molecular complex that generates NO which acts as a signaling molecule and has antimicrobial properties. iNOS activity is induced during immune responses and contributes to the elimination of bacteria and viruses through the production of reactive nitrogen species. Its expression is tightly regulated by cytokines and other signaling molecules that modulate the extent of its action.
Pathways
INOS integrates into the nitric oxide signaling pathway and is closely associated with inflammatory pathways. NO serves as a messenger within the nitric oxide signaling pathway facilitating processes like vasodilation and neurotransmission. iNOS works alongside other proteins such as cytokines like interferon-gamma that elevate its activity during inflammation and immune responses. The regulation of iNOS expression and activity plays an essential role in maintaining immune homeostasis and ensuring proper response to infections.
