Mouse Interferon alpha 2 ELISA Kit (ab211648) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Interferon alpha 2 protein in cell culture extracts, cell culture supernatant, plasma, serum, and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse Interferon alpha 2 with 0.9 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. A 384-well SimpleStep ELISA® microplate (Pre-coated 384 well Microplate SimpleStep ELISA® ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
There are three major structural types of interferons: alpha, beta, and gamma. Interferon alpha forms are produced by monocytes/macrophages, fibroblasts, and a number of different cell types following induction by viruses, nucleic acids, and glucocorticoid hormones. Interferon alpha-2 is one of at least 23 different known variants of interferon alpha. The individual interferon alpha proteins have molecular masses between 19-26 kDa and consist of proteins with lengths of 156 - 172 amino acids (aa). Mouse Interferon alpha-2 is 167 aa in length. Interferon alpha-2 demonstrates antiviral, antiparasitic, and antiproliferative activities.

Interferon alpha 2 also known as IFN alpha 2 or IFN alpha 2a/b is a type of cytokine that plays a critical role in the immune response. It is a smaller protein weighing approximately 19.5 kDa mainly expressed by leukocytes such as macrophages and dendritic cells. This protein exhibits antiviral antiproliferative and immunomodulatory activities by binding to specific receptors on target cells. Researchers often study interferon alpha through recombinant protein studies which can be simulated by companies like PeproTech. Additionally ELISA assays facilitate the quantitative measurement of interferon alpha levels in various biological samples.
Interferon alpha 2 engages in complex signaling with other immune factors to establish an antiviral state in cells. This protein binds to the IFNAR1 and IFNAR2 receptor complex initiating a cascade that activates genes responsible for immune defense. By doing so it triggers the transcription of various antiviral proteins and enhances the activity of immune cells including natural killer cells and macrophages. Its function is also evaluated in reconstitution studies such as PeproTech's IL-2 reconstitution to investigate broader immune functions.
Researchers observe interferon alpha 2 predominantly within the JAK-STAT signaling pathway. This pathway is vital in transmitting the extracellular signal through to the gene transcription process leading to an appropriate immune response. STAT proteins such as STAT1 and STAT2 form a transcription complex following activation by JAK kinases. Interferon alpha 2's interactions in the pathway exemplify how this cytokine links with other components for immune regulation coordinating with other proteins like ISGF3 to amplify antiviral responses.
Scientists associate interferon alpha 2 with chronic viral infections and certain cancers. For example its therapeutic application has been prominent in treating hepatitis C and some hematological malignancies. In viral infections interferon alpha inhibits viral replication and enhances antiviral immunity. In cancer it suppresses cell proliferation and regulates immune surveillance. The protein's levels and activity can correlate with pathological states influenced by other proteins such as IFN-lambda and TNF-alpha highlighting its role in disease mechanisms and therapeutic interventions.

The concentrations of Interferon alpha-2 were measured in duplicates, interpolated from the Interferon alpha-2 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 100%, plasma (citrate) 100%, plasma (EDTA) 100%, and plasma (heparin) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Interferon alpha-2 concentration was determined to be 196.9 pg/mL in serum, 181.9 pg/mL in plasma (citrate), 204.1 pg/mL in plasma (EDTA), and 194.9 pg/mL in plasma (heparin).

The concentrations of Interferon alpha-2 were measured in duplicates, interpolated from the Interferon alpha-2 standard curves and corrected for sample dilution. Undiluted samples are as follows: 100% RAW 264.7 stimulated supernatant, 100% J774A.1 stimulated supernatant, and 100% PBMC stimulated supernatant. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Interferon alpha-2 concentration was determined to be 466.4 pg/mL in spiked RAW 264.7 stimulated supernatant, 500.1 pg/mL in spiked J774A.1 stimulated supernatant, and 518.9 pg/mL in spiked PBMC stimulated supernatant. RAW 264.7 supernatant was stimulated for 48 hours with 5 μg/mL of LPS. J774A.1 supernatant was stimulated for 72 hours with 1.5% PHA and 10 ng/mL PMA. PBMC supernatant was stimulated for 5 hours with 3 μg/mL of imiquimod.
