Sandwich ELISA - Mouse JAM-A ELISA Kit (Junctional Adhesion Molecule) (AB277080)
Interpolated concentrations of native JAM-A in mouse placenta based on a 25 µg/mL extract load, lung based on a 25 µg/mL extract load, and spleen based on a 50 µg/mL extract load.
The concentrations of JAM-A were measured in duplicate and interpolated from the JAM-A standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean JAM-A concentration was determined to be 1159.97 pg/mL in placenta extract, 2151.45 pg/mL in lung extract, and 1030.16 pg/mL in spleen extract.

Sandwich ELISA - Mouse JAM-A ELISA Kit (Junctional Adhesion Molecule) (AB277080)
Interpolated concentrations of native JAM-A in mouse serum and plasma samples.
The concentrations of JAM-A were measured in duplicates, interpolated from the JAM-A standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma (citrate) 50%, plasma (EDTA) 50%, and plasma (heparin) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean JAM-A concentration was determined to be 2571.47 pg/mL in serum, 1804.58 pg/mL in plasma (citrate), 1825.22 pg/mL in plasma (EDTA), and 2347.57 pg/mL in plasma (heparin).

Product details
Mouse JAM-A ELISA kit (ab277080) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Mouse JAM-A protein in mouse serum, plasma, cell culture media, and cell and tissue extract. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse JAM-A with 2.92 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Junction Adhesion Molecule (JAM-A), a type I transmembrane protein, appears to play a role in epithelial tight junction formation. JAM-A emerges early in primordial forms of cell junctions and recruits PARD3. The association of the PARD6-PARD3 complex may prevent the interaction of PARD3 with JAM-A, thereby preventing tight junction assembly. JAM-A plays a role in regulating monocyte transmigration involved in integrity of epithelial barrier. JAM-A shares 68.4%, 91.3%, 96.3%, and 70.1% sequence homology with human, rat, bovine, and monkey, respectively.

Biological function summary
JAM-A functions as a cell adhesion molecule influencing processes like leukocyte transmigration and epithelial barrier formation. It does not act alone but rather forms complexes with other proteins at cell junctions. JAM-A interacts with other junctional proteins like occludin and claudins contributing to the structure and function of tight junctions.
Pathways
JAM-A participates in cellular processes involving inflammation and cell migration. It is an integral part of the signal transduction pathways that regulate leukocyte migration an important aspect of immune response. JAM-A is also associated with G-protein coupled receptors in these pathways highlighting its role in regulating intracellular signaling that affects cell adhesion and migration.
