Mouse LIX ELISA Kit (CXCL5) (ab264611) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of LIX (CXCL5) protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, serum, urine, and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse LIX (CXCL5) with 1.14 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies

Mouse LIX (LPS-induced CXC chemokine) also known as CXCL5, is a member of the ELR+ group of 7-9 kDa neutrophil and monocyte chemotactic proteins in the CXCL cytokine family. Mouse LIX can be produced by fibroblasts, epithelial cells (including alveolar type II epithelia), endothelial cells, platelets, cardiac myocytes, preosteoblasts, oligodendrocytes, and adipose-resident macrophages. It is mainly produced when induced by LPS, IL-17 and/or TNF-α. Endotoxemia increases LIX expression, especially in the heart, but also in the lung, spleen, and liver. LIX is downregulated by glucocorticoids and is considered a glucocorticoid-attenuated response gene or GARG. It can also be downregulated by IL-10 and viral proteins. LIX is the most potent mouse neutrophil chemoattractant and activator LIX participates in the induction of LPS-induced acute lung injury and in lung ischemia-induced angiogenesis. High expression of LIX in obese white adipose tissue is thought to contribute to insulin resistance.Mouse CXCL5 is the equivalent of human CXCL6 based on genetic and biological evidence as pointed out by Nomiyama *et al.* (PMID: 20434943).

Activity summary

CXCL5 also known as LIX (Liposaccharide-induced CXC chemokine) is a small cytokine belonging to the CXC chemokine family. It plays a significant role in immunological responses by acting as a chemoattractant for neutrophils. CXCL5 has a molecular mass of around 12 kDa. The protein is mainly expressed in neutrophils epithelial cells and endothelial cells. It is often studied alongside its competitors in different therapeutic and diagnostic contexts.

Biological function summary

CXCL5 affects inflammation and the immune response by modulating neutrophil recruitment to sites of infection or injury. It is a part of the larger network of chemokines that direct leukocyte movement throughout the body. While CXCL5 does not form a complex in a traditional sense it does interact dynamically with CXCR2 receptor to exert its effects. These interactions are pivotal to the protein’s role in orchestrating the immune system’s response to diverse stimuli.

Pathways

CXCL5 plays a part in the chemokine signaling pathway and the inflammatory response pathway. These pathways are important in mediating immune surveillance and inflammation. CXCL5 through these pathways has a strong connection to other CXC family proteins like CXCL1 and the receptor CXCR2 which allows for a coordinated response during immune challenges and recovery processes.

Associated diseases and disorders

CXCL5 is relevant in the context of chronic inflammatory disorders such as rheumatoid arthritis and cancer specifically its role in tumor-related inflammation. In these conditions CXCL5 is linked to the exacerbation of inflammatory states and promotion of tumor growth via angiogenesis. The protein's interactions with CXCR2 play a significant role in these disease processes representing potential therapeutic targets for managing inflammation and modulating cancer progression.
