Mouse MCP1 ELISA Kit (ab208979) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of MCP1 protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse MCP1 with 0.487 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. MCP1 (CCL2) is a chemokine expressed by fibroblasts, macrophages, endothelial cells, mast cells, osteoblasts, ameloblasts, epithelial cells, and astrocytes after stimulation with inflammatory stimuli. MCP1 is involved in the recruitment of monocytes and lymphocytes and in the regulation of Th1/Th2 lymphocyte differentiation.The standard is affinity purified.
MCP-1 also known as CCL2 is a chemokine involved in the recruitment of monocytes to sites of injury and inflammation. This protein consists of 76 amino acids and has a molecular weight of about 11 kDa. MCP-1 is expressed in a wide range of cell types including endothelial cells fibroblasts and monocytes. It acts by binding to the chemokine receptor CCR2 stimulating intracellular signaling pathways that mediate cell migration.
The role of MCP-1 is significant in guiding immune cells to areas requiring immune response. It does not form part of a complex but operates as a singular entity to affect immune signaling. MCP-1 influences the behavior of monocytes and macrophages by inducing chemotaxis and activating their effector functions. Through this mechanism it aids in the inflammatory response and tissue repair.
MCP-1 participates in the inflammatory pathways by cooperating with key proteins like CCR2. It facilitates the JAK-STAT signaling pathway which is vital for transmitting extracellular information into cellular responses. The interaction of MCP-1 with CCR2 influences the activation of downstream proteins like STAT3 enabling various immune responses. MCP-1 thereby plays a central role in modulating inflammation-related pathways.
MCP-1 has a connection to inflammatory bowel disease and atherosclerosis. In such conditions elevated levels of MCP-1 can exacerbate inflammatory responses by recruiting monocytes to dysfunctional sites worsening tissue damage. Through its interaction with CCR2 MCP-1 becomes a pivotal factor in chronic inflammation. Targeting this MCP-1/CCR2 axis can provide therapeutic opportunities for modulating disease progression in these inflammatory diseases.

Standard curve comparison between mouse MCP1 SimpleStep ELISA® kit and traditional ELISA kit from leading competitor. SimpleStep ELISA kit shows a 3-fold increase in sensitivity.
Interpolated concentrations of native MCP1 in mouse serum, plasma and cell culture supernatant samples.
The concentrations of MCP1 were measured in duplicates, interpolated from the MCP1 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 25%, plasma (citrate) 25%, plasma (EDTA) 25%, and RAW264.7 cell culture supernatant 10%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MCP1 concentration was determined to be 157.7 pg/mL in serum, 125.7 pg/mL in plasma (citrate), 121.1 pg/mL in plasma (EDTA), and 277.2 pg/mL in RAW264.7 cell culture supernatant.
