Sandwich ELISA - Mouse MMP9 ELISA Kit (AB253227)
Interpolated concentrations of native MMP9 in mouse J774A.1 cell culture supernatant samples.
The concentrations of MMP9 were measured in duplicates, interpolated from the MMP9 standard curves and corrected for sample dilution. Undiluted samples are as follows : J774A.1 cell culture supernatant 0.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MMP9 concentration was determined to be 1326.73 ng/mL in J774A.1 cell culture supernatant.

Sandwich ELISA - Mouse MMP9 ELISA Kit (AB253227)
Interpolated concentrations of native MMP9 in mouse LPS unstimulated and stimulated P388D1 cell culture supernatant samples.
The concentrations of MMP9 were measured in duplicates, interpolated from the MMP9 standard curves and corrected for sample dilution. Undiluted samples are as follows : unstimulated P388D1 supernatant 100% and LPS stimulated P388 D1 10%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MMP9 concentration was determined to be 1.03 ng/mL in unstimulated P388D1 supernatant and 74.58 ng/mL in LPS stimulated P388D1.

Interpolated concentrations of native MMP9 in mouse serum and plasma - heparin samples.
The concentrations of MMP9 were measured in duplicates, interpolated from the MMP9 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 5% and plasma (heparin) 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MMP9 concentration was determined to be 158.72 ng/mL in serum and 37.15 ng/mL in plasma (heparin).

Product details
Mouse MMP9 ELISA Kit ab253227 is a rapid single-wash 90-min Sandwich ELISA to measure Mouse MMP9 in cell culture supernatant, heparin plasma, serum, tissue extracts. This SimpleStep sensitivity is 38.91 pg/mL.
How the assay works
Mouse MMP9 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Assay Specificity
Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
Please refer to our protocol booklet for more details.
Mouse MMP9 ELISA Kit ab253227 protocol summary

MMP9 (gelatinase B) is a member of the matrix metalloproteinases family of zinc and calcium dependent endopeptidases. MMP9 is a 95kDa protein produced by keratinocytes, monocytes, macrophages and PMN leukocytes. It is able to degrade all the components of the extracellular matrix including gelatin, collagen types IV and V, elastin and proteoglycan core protein. MMP9 is believed to act synergistically with interstitial collagenase (MMP-1) in the degradation of fibrillar collagens as it degrades their denatured gelatin forms. MMP9 is present in most inflammatory responses. Mouse MMP9 shares 72.9% sequence homology with primate. 71.8% with human, 81.6 % with rat, and 73.0% with bovine MMP9.

Biological function summary
Matrix metalloproteinase-9 profoundly influences tissue remodeling inflammation and angiogenesis. It does not form part of a larger protein complex but interacts closely with substrates in the extracellular matrix. MMP-9 plays important roles in processes such as embryogenesis reproduction and wound healing through the remodeling of surrounding tissues. The MMP-9 protein is often measured using assays like ELISA or analyzed via techniques such as Western blotting to determine its expression levels and activity in different biological contexts.
Pathways
MMP-9 participates in the matrix metalloproteinase pathway and the NF-kB signaling pathway. These pathways are essential in orchestrating processes such as tissue remodeling and inflammatory responses. MMP-9 acts alongside other matrix metalloproteinases including MMP-2 and is regulated by factors like cytokines growth factors and hormones ensuring the precision of extracellular matrix degradation during physiological processes.
