Sandwich ELISA - Mouse Myeloperoxidase ELISA Kit (MPO) (AB275109)
Interpolated concentrations of native Myeloperoxidase in mouse serum and plasma samples.
The concentrations of Myeloperoxidase were measured in duplicates, interpolated from the Myeloperoxidase standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 7.5%, plasma (citrate) 3.8%, plasma (EDTA) 7.5% and plasma (heparin) 7.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Myeloperoxidase concentration was determined to be 138 ng/mL in neat serum, 244 ng/mL in plasma (citrate) and 124 ng/mL in neat plasma (EDTA) and 123 ng/mL in neat plasma (heparin).

Product details
Mouse Myeloperoxidase ELISA Kit (MPO) ab275109 is a rapid single-wash 90-min Sandwich ELISA to measure Mouse Myeloperoxidase (MPO)in cell culture media, citrate plasma, EDTA plasma, heparin plasma, serum. This SimpleStep sensitivity is 117 pg/mL.
How the assay works
Mouse Myeloperoxidase (MPO)SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Assay Specificity
Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
Please refer to our protocol booklet for more details.

2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm
Myeloperoxidase (MPO) is part of the host defense system of polymorphonuclear leukocytes. It is responsible for microbicidal activity against a wide range of organisms. MPO is a hemoprotein expressed in the neutrophils (polymorphonuclear leukocytes) and is secreted during activation. Myeloperoxidase catalyzes the oxidation of chloride ions to hypochlorus acid, which is a potent antimicrobial agent. Myeloperoxidase pro- oxidative and pro-inflammatory properties cause an increase in MPO in multiple diseases including heart disease, myocardial infraction, and multiple sclerosis. Myeloperoxidase is a clinical marker of myocardial infraction and cardiovascular disease.

Biological function summary
The generation of reactive oxygen species by myeloperoxidase is essential for microbicidal activity. Myeloperoxidase functions as part of the antimicrobial system in the phagosome which is the intracellular compartment where pathogens are degraded. This enzyme works in conjunction with other components of the immune system such as NADPH oxidase. By generating hypochlorous acid MPO contributes to the oxidative burst a rapid release of reactive oxygen species during the response to pathogens.
Pathways
Myeloperoxidase integrates into the immune defense and inflammatory pathways. In particular it is associated with the neutrophil degranulation pathway where it releases its enzymatic contents to fight off microbes. MPO also interacts with proteins involved in oxidative stress processes such as superoxide dismutase which moderates levels of reactive oxygen species in cells. These interactions ensure balance in the immune response preventing excessive tissue damage during inflammation.
