Sandwich ELISA - Mouse Nestin ELISA Kit (AB223859)
Interpolated concentrations of native Nestin in Mouse NIH/3T3 cell extract sample and sample based on a 62.5 μg/mL extract load.
The concentrations of Nestin were measured in duplicate and interpolated from the Nestin standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Nestin concentration was determined to be 678 pg/mL in NIH/3T3 Cell extract.

Product details
Mouse Nestin ELISA Kit (ab223859) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Nestin protein in cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse Nestin with 7.6 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Nestin is an intermediate filament protein and is a marker for neural stem cells. Nestin is required for brain and eye development. Nestin expression generally doesn't persist into adult hood, with the exception of the neuronal precursor cells of the subgranular zone. After cells have differentiated Nestin is downregulated and is replaced by tissue-specific intermediate filament proteins.

Biological function summary
Nestin plays a significant role in the dynamic remodeling of the cytoskeleton. It does not act as a solitary element but forms part of a larger protein complex involved in maintaining and organizing cellular structure. This property grants cells flexibility in shape and resilience during the rapid changes in development and specialization. The role of Nestin in the cellular architecture allows it to contribute to processes such as cell migration and proliferation critical during tissue development and repair.
Pathways
Nestin integrates into networks that regulate stem cell behavior and neurogenesis. It tightly interacts within the pathways involving cell cycle regulation and differentiation. For example the Wnt and Notch signaling pathways modulate its expression to influence neural stem cell fate and patterning during development. Protein interactions between Nestin and other cytoskeletal elements like vimentin facilitate its roles in these pathways adapting the cellular environment for specific developmental activities.
