Function

Component of N-methyl-D-aspartate (NMDA) receptors (NMDARs) that function as heterotetrameric, ligand-gated cation channels with high calcium permeability and voltage-dependent block by Mg(2+) (PubMed:12008020, PubMed:12860921, PubMed:14602821, PubMed:1532151, PubMed:8060614, PubMed:9049108). NMDARs participate in synaptic plasticity for learning and memory formation by contributing to the long-term potentiation (LTP) (By similarity). Channel activation requires binding of the neurotransmitter L-glutamate to the GluN2 subunit, glycine or D-serine binding to the GluN1 subunit, plus membrane depolarization to eliminate channel inhibition by Mg(2+) (PubMed:12008020, PubMed:1532151, PubMed:7790891, PubMed:8060614). NMDARs mediate simultaneously the potasium efflux and the influx of calcium and sodium (PubMed:12860921). Each GluN2 or GluN3 subunit confers differential attributes to channel properties, including activation, deactivation and desensitization kinetics, pH sensitivity, Ca2(+) permeability, and binding to allosteric modulators (PubMed:12008020, PubMed:14602821).

Mouse NMDAR1 ELISA kit (ab282862) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Mouse NMDAR1 protein in mouse cell and tissue extract samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse NMDAR1 with 27.74 pg/mL sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Mouse NMDA receptor (NMDAR1) is a and protein receptor activated upon and glutamate binding. NMDAR1 is a component of NMDA receptor complexes that function as heterotetrameric, ligand-gated ion channels with high calcium permeability and voltage-dependent sensitivity to magnesium. NMDA receptor complex channel activation requires binding of the neurotransmitter glutamate to the epsilon subunit, glycine binding to the zeta subunit, plus membrane depolarization to eliminate channel inhibition. Mouse NMDAR1 shares 99.8% and 99.3% sequence homology with rat and human, respectively.

Activity summary

NMDAR1 also known as GluN1 is a subunit of the NMDA receptor a type of ion channel protein found in the brain. This receptor weighs around 120 kDa and plays an important role in synaptic transmission and plasticity. NMDAR1 operates as part of a receptor complex that allows for calcium influx when activated. It shows high expression throughout the central nervous system primarily in neurons across the cortex and hippocampus. The receptor's function is important to certain calcium-dependent processes in the brain.

NMDA receptors including the NMDAR1 subunit serve essential functions in synaptic plasticity and memory processes. As part of the receptor complex NMDAR1 contributes to the control of synaptic strength. NMDA receptor dysfunction including the NMDAR1 subunit links to neurodegenerative diseases such as Alzheimer's. The alterations in NMDAR1 function can lead to excitotoxicity contributing to neuronal damage. NMDAR1 also associates with schizophrenia where altered glutamatergic transmission has been observed. Here abnormalities in proteins like the synaptic scaffolding protein PSD-95 have also been noted to relate to NMDAR1 dysfunction in the pathophysiology of these disorders.

    Interpolated concentrations of native NMDAR1 in mouse brain tissue extract based on a 125 μg/mL extract load.

    The concentrations of NMDAR1 were measured in duplicate and interpolated from the NMDAR1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean NMDAR1 concentration was determined to be 7515.05 pg/mL in brain tissue extract.
