This assay detects rat p27Kip1.

The concentrations of p27Kip1 in rat brain extract were measured in duplicate, interpolated from the mouse p27Kip1 standard curves and corrected for sample dilution. Undiluted samples are rat brain extract 62.5 μg/mL. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean p27Kip1 concentration was determined to be 1,077 pg/mL in rat brain extract.

The concentrations of p27Kip1 were measured in duplicate and interpolated from the p27Kip1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean p27Kip1 concentration was determined to be 496 pg/mL in NIH3T3 extract and 634 pg/mL in mouse skeletal muscle extract.

Mouse p27Kip1 ELISA Kit (ab208982) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of p27Kip1 protein in cell culture extracts and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse p27Kip1 with 7.5 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

- Single-wash protocol reduces assay time to 90 minutes or less

Cyclin-dependent kinase inhibitor 1B (p27Kip1) is an enzyme inhibitor that is part of the Cip/Kip family of cyclin dependent kinase inhibitor proteins. p27Kip1 is a regulator of cell cycle progression involved at the G1 stage by binding to and preventing the activation of cyclin E-CDK2 or cyclin D-CDK4 complexes.

The protein known as p27 KIP1 also referred to as p27 CDKN1B or KIP1 is a member of the kinase inhibitory protein family. It plays an important role in cell cycle regulation by inhibiting cyclin-dependent kinases (CDKs). p27 KIP1 weighs approximately 27 kDa and can be found in various tissues where it regulates cell proliferation. The protein functions as a suppressor by binding to cyclin-CDK complexes preventing the transition from G1 phase to S phase in the cell division cycle.
Biological function summary

The function of p27 KIP1 involves its role in controlling cell growth and division. It achieves this by becoming a part of larger protein complexes involving CDKs and cyclins. By directly interacting with these complexes p27 KIP1 modulates cell cycle progression therefore acting as a brake on cellular proliferation.
