Interpolated concentrations of native PD-1H in mouse spleen extract, lung extract, placenta extract, and RAW 264.7 cell extract.
The concentrations of PD-1H were measured in duplicate and interpolated from the PD-1H standard curve and corrected for sample dilution. Undiluted samples are 250 μg/mL for all extracts. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

Interpolated concentrations of native PD-1H in mouse serum and plasma samples.
The concentrations of PD-1H were measured in duplicates, interpolated from the PD-1H standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma (citrate) 50%, plasma (EDTA) 100%, and plasma (heparin) 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PD-1H concentration in the neat samples was determined to be 19.7 ng/mL in serum, 17.8 ng/mL in plasma (citrate), 15.3 ng/mL in plasma (EDTA), and 31.8 ng/mL in plasma (heparin).

Mouse PD-1H ELISA Kit (ab231933) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of PD-1H protein in cit plasma, edta plasma, hep plasma, serum, tissue extracts, and cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse PD-1H with 44.3 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

Biological function summary
VISTA functions in immune checkpoint regulation by modulating T cell activity. It acts as both a ligand and a receptor influencing immune responses. As part of an immune complex VISTA interacts with other checkpoint proteins to maintain immune homeostasis and prevent autoimmunity. Its role mirrors those of other B7 family members like PD-1 and CTLA-4 which are involved in negative immune regulation.
Pathways
VISTA integrates into the broader framework of immune signaling pathways. It contributes significantly to the immune checkpoint pathway similar to CTLA-4 and PD-1 which mediate immune cell proliferation and activation. By interacting with these proteins VISTA plays an important role in maintaining self-tolerance and regulating inflammatory responses functioning parallel to other proteins such as CD28 and B7-H5 in the immune checkpoint pathway.
