Mouse PD-L1 ELISA Kit- Intracellular is a single-wash 90-min Simplestep used to quantify Mouse PD-L1 with a sensitivity of 7.057 pg/mL. The assay uses a simple mix-wash-read protocol with just one incubation and one wash step.

Interpolated concentration of native PD-L1 was measured in duplicate at different sample concentrations. Undiluted samples are as follows : LPS treated RAW264.6 cell extract 125 μg/mL (treated with 1 µg/ml LPS for 48 hrs), mouse lung tissue extract 250 μg/mL, and mouse lymph node tissue extract 250 μg/mL. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer SSW.

Mouse PD-L1 SimpleStep ELISA® kit is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of PD-L1 protein in Cell Lysate, Tissue extracts. Quantitate Mouse PD-L1 with 7.057 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips, available in 10-pack (10 x 96-well plates)

Target data

Plays a critical role in induction and maintenance of immune tolerance to self (PubMed : 11238124). As a ligand for the inhibitory receptor PDCD1/PD-1, modulates the activation threshold of T-cells and limits T-cell effector response (PubMed : 11238124). Through a yet unknown activating receptor, may costimulate T-cell subsets that predominantly produce interleukin-10 (IL10) (PubMed : 11015443, PubMed : 12719480).. The PDCD1-mediated inhibitory pathway is exploited by tumors to attenuate anti-tumor immunity and escape destruction by the immune system, thereby facilitating tumor survival (PubMed : 12218188, PubMed : 27281199). The interaction with PDCD1/PD-1 inhibits cytotoxic T lymphocytes (CTLs) effector function (PubMed : 12218188). The blockage of the PDCD1-mediated pathway results in the reversal of the exhausted T-cell phenotype and the normalization of the anti-tumor response, providing a rationale for cancer immunotherapy (PubMed : 12218188).
