Interpolated concentrations of native PD-L2 in mouse serum, plasma, and urine samples.
The concentrations of PD-L2 were measured in duplicates, interpolated from the PD-L2 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma (citrate) 100%, plasma (EDTA) 100%, plasma (heparin) 100% and urine 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PD-L2 concentration was determined to be 699.03 pg/mL in serum, 272.41 pg/mL in plasma (citrate), 310.34 pg/mL in plasma (EDTA), 412.41 pg/mL in plasma (heparin) and 47.99 pg/mL in urine.

Mouse PD-L2 ELISA Kit (ab283880) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of PD-L2 protein in serum, plasma and urine samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse PD-L2 with 1.03 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less

Mouse PD-L2 is involved in the costimulatory signal, essential for T-cell proliferation and IFNG production in a PDCD1-independent manner. Interaction with PDCD1 inhibits T-cell proliferation by blocking cell cycle progression and cytokine production.

Biological function summary
PD-L2 offers protection to tissues from autoimmunity by regulating T-cell activation and fostering tolerance. By engaging with its receptor PD-1 PD-L2 forms a complex that modulates the immune response to prevent overactivation. This interaction limits the immune system's potential damage to healthy tissues maintaining proper immune function in diverse environments.
Pathways
PD-L2 participates in the immune checkpoint pathways where it works closely with PD-1 to mediate immune responses. PD-L2 forms an important part of the program death-1 (PD-1) pathway which plays a prominent role in downregulating T-cell activity. It relates to proteins such as PD-L1 which also interacts with PD-1 modulating immune tolerance and response to infections or tumors.
