Collagen type III occurs in most soft connective tissues along with type I collagen. Involved in regulation of cortical development. Is the major ligand of ADGRG1 in the developing brain and binding to ADGRG1 inhibits neuronal migration and activates the RhoA pathway by coupling ADGRG1 to GNA13 and possibly GNA12.

Procollagen III N-terminal propeptide known as PIIINP serves as an important marker in collagen synthesis processes. PIIINP results from the cleavage of procollagen III and functions as a byproduct of the maturation of collagen molecules. It possesses a molecular weight of approximately 42 kDa. This target is initially expressed within fibroblast cells found in connective tissues and then secreted into the bloodstream. Given its role in collagen synthesis it is frequently measured to assess the turnover and formation of collagen in tissues.
This propeptide plays a significant role in the formation of the extracellular matrix. PIIINP is not part of a protein complex but accumulates during the conversion of procollagen into mature collagen fibers which later support tissue structure. Its presence is essential for proper collagen deposition and tissue remodeling. Through its involvement in collagen synthesis PIIINP helps maintain the structural integrity of connective tissues across various organs.
Its role reflects the broader functions of collagen metabolism and tissue remodeling pathways. PIIINP marks the processing pathway of procollagen to collagen which is important for the formation of the extracellular matrix. This pathway sometimes intersects with pathways involving other collagens such as collagen I which is another critical component of connective tissues. Together PIIINP and collagen I contribute to the proper maintenance of the extracellular matrix and the mechanical properties of tissues.
Elevated levels of PIIINP often relate to fibrotic conditions and can serve as a biomarker for diseases like liver fibrosis and systemic sclerosis. In liver fibrosis excessive collagen deposition and remodeling occur affecting liver function. In systemic sclerosis abnormal collagen synthesis and deposition in the skin cause thickening and hardening. PIIINP connection to these conditions often involves collagen-binding proteins which regulate collagen stability and deposition.
