Interpolated concentrations of native PTX3 in mouse serum, plasma, and cell culture supernatant samples. The concentrations of PTX3 were measured in duplicates, interpolated from the PTX3 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 25%, plasma (citrate) 12.5%, plasma (EDTA) 12.5%, plasma (heparin) 12.5% and 3T3-L1 cell culture supernatant 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PTX3 concentration was determined to be 36.40 ng/mL in serum, 33.63 ng/mL in plasma (citrate), 50.51 ng/mL in plasma (EDTA), 51.03 ng/mL in plasma (heparin), and 8.37 ng/mL in 3T3-L1 cell culture supernatant.

Mouse PTX3 ELISA Kit (ab245713) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of PTX3 protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse PTX3 with 22.3 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Biological function summary
The role of PTX3 extends beyond simple pathogen recognition. It is involved in the regulation of inflammation and tissue homeostasis. As part of the pentraxin superfamily PTX3 plays a role in the formation of complexes with other molecules such as complement components. These interactions modulate the complement cascade an essential part of the innate immune response and influence the resolution of inflammation. PTX3 also interacts with extracellular matrix components stabilizing tissue structure during immune response.
Pathways
PTX3 integrates into vital immune signaling networks. It actively participates in the complement pathway where it regulates the activity of C1q and C3 key complement proteins. Additionally PTX3 connects with the Toll-like receptor (TLR) signaling pathway. This interaction helps modulate the body's response to microbial infections by influencing the activity of TLR4 among other related proteins which are fundamental in recognizing pathogen-associated molecular patterns.
