Product details

This Mouse/rat Prolactin kit (ab285344, K4688) is a solid phase sandwich ELISA Kit. The samples and biotinylated anti-prolactin antibodies are added to the wells coated with polyclonal antibody to prolactin. Prolactin in samples binds to the anti-prolactin polyclonal antibody on the well and the biotinylated anti-prolactin antibody binds to prolactin. Unbound protein and biotin conjugate are washed off by wash buffer. Horseradish peroxidase conjugated streptavidin is added to each wells and the streptavidin is allowed to bind to the biotin in the complex. Unbound enzyme conjugate is washed off. Upon the addition of the substrate, the intensity of color is proportional to the concentration of prolactin in the samples. A standard curve is prepared relating color intensity to the concentration of the prolactin.

Supplementary information

Prolactin also known as PRL is a protein hormone with a molecular mass of approximately 23 kDa. It is primarily produced in the anterior pituitary gland but is also found in other tissues such as the placenta mammary glands and immune cells. Mechanically Prolactin functions as a peptide hormone and exerts its effects by binding to the Prolactin receptor a member of the cytokine receptor superfamily. This binding activates the Jak-STAT signaling pathway which is important for mediating the hormone's effects in target cells.
Biological function summary

The functions of this hormone extend beyond its well-known role in lactation. It helps stimulate milk production in the mammary glands by promoting the proliferation and differentiation of mammary epithelial cells. Additionally Prolactin influences reproductive functions metabolism and immune regulation. It is not part of a larger protein complex but interacts with various proteins to perform its functions. Notably it modulates the immune response contributing to tolerance during pregnancy and affecting the balance between pro-inflammatory and anti-inflammatory cytokines.

Pathways

Prolactin plays important roles in the Jak-STAT and MAPK signaling pathways. In these pathways it shares interactions with related proteins such as Growth Hormone and Prolactin Receptor aiding in cell growth differentiation and metabolism regulation. Prolactin's involvement in these pathways highlights its central role in physiological processes linking various hormonal signals to regulate body functions efficiently.

Prolactin's imbalance can lead to conditions such as hyperprolactinemia and prolactinomas both characterized by excessive levels of this hormone in the blood. These conditions are often related to alterations in the pituitary gland function. Prolactin interacts closely with Dopamine a neurotransmitter that inhibits its secretion influencing both its production and release. Dysregulation of this interaction may contribute to the development of these disorders impacting endocrine health and overall well-being.
