Interpolated concentrations of native Visfatin in mouse and rat, cell and tissue extracts, based on a 25 μg/mL extract load.

The concentrations of Visfatin were measured in duplicate and interpolated from the Visfatin standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Visfatin concentration was determined to be 12,513.20 pg/mL in Raw 264.7 cell extract, 9,298.50 pg/mL in mouse liver tissue extract, 10,138.83 pg/mL in C6 cell extract, and 13,465.27 pg/mL in rat liver tissue extract.

Mouse/Rat Visfatin ELISA Kit (ab267799) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Visfatin protein in cell lysate and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse/Rat Visfatin with 167.5 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips

A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Visfatin, also known as nicotinamide phosphoribosyltransferase, is the key enzyme in the salvage pathway by which the cell converts nicotinamide to nicotinamide mononucleotide. Along with its enzymatic role, Visfatin is thought to play a pivotal role in B-cell maturation and cell signaling. This protein is highly conserved throughout Mammalia, subsequently, this kit cross-reacts with human.

The secreted form behaves both as a cytokine with immunomodulating properties and an adipokine with anti-diabetic properties, it has no enzymatic activity, partly because of lack of activation by ATP, which has a low level in extracellular space and plasma (By similarity). Catalyzes the condensation of nicotinamide with 5-phosphoribosyl-1-pyrophosphate to yield nicotinamide mononucleotide, an intermediate in the biosynthesis of NAD. It is the rate limiting component in the mammalian NAD biosynthesis pathway. Plays a role in the modulation of circadian clock function. NAMPT-dependent oscillatory production of NAD regulates oscillation of clock target gene expression by releasing the core clock component : CLOCK-BMAL1 heterodimer from NAD-dependent SIRT1-mediated suppression.
