The Mouse Relaxin 1 Enzyme-Linked Immunosorbent Assay (ELISA) kit (ab213885) is designed for the quantitative measurement of Mouse Relaxin 1 in cell culture supernatants, serum and plasma (heparin or EDTA ).
The ELISA kit is based on standard sandwich enzyme-linked immunosorbent assay technology. A monoclonal antibody from rat specific for Relaxin 1 has been pre-coated onto 96-well plates. Standards (Expression system for standard: E.coli; Immunogen sequence: R23-R54 (B) & E161-C185 (A)) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Relaxin 1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Mouse Relaxin 1 amount of sample captured in plate.
Relaxin 1 is a member of relaxin-like peptide family. Relaxin gene maps to mouse chromosome 19 near D19Mit23. Relaxin is a peptide hormone produced by the corpora lutea of ovaries during pregnancy in many mammalian species, including man. Relaxin widens the pubic bone and facilitates labor, it also softens the cervix (cervical ripening), and relaxes the uterine musculature. However, its significance may reach much further. Relaxin affects collagen metabolism, inhibiting collagen synthesis and enhancing its breakdown by increasing matrix metalloproteinases. It also enhances angiogenesis and is a potent renal vasodilator.
Relaxin 1 also known as RLN1 is a peptide hormone that plays a role in reproductive biology. It has a mass of approximately 6 kilodaltons. Relaxin 1 is expressed in various tissues mainly the reproductive system including the ovaries placenta and uterus. It modulates connective tissue remodeling which is important during childbirth by helping to relax the pelvic ligaments. Additionally it has effects on the cardiovascular and nervous systems.
The hormone influences reproductive processes through its action on tissues affecting aspects like collagen remodeling and vasodilation. It does not form part of a complex but interacts with receptors such as RXFP1 which are G-protein-coupled receptors. The binding of Relaxin 1 to RXFP1 receptors activates several downstream signaling pathways that facilitate its biological functions.
The role of Relaxin 1 extends beyond reproductive functions. It is involved in the extracellular matrix pathway and can influence the nitric oxide signaling pathway which impacts vascular relaxation and blood flow. Relaxin 1 related proteins include RXFP1 and its family member Relaxin 2 which can bind to the same receptor and trigger similar effects.
Relaxin 1 has a connection to fibrotic diseases and cardiovascular conditions. It exhibits antifibrotic properties reducing tissue scarring in organs like the skin and lungs. In cardiovascular diseases Relaxin 1 can help improve heart functions through its vasorelaxation effects. In these disease contexts Relaxin 1 interacts with proteins involved in the extracellular matrix and nitric oxide pathways to mediate its impact.
