Sandwich ELISA - Mouse SET7 ELISA Kit (AB279404)
Interpolated concentrations of native SET7 in mouse NIH-3T3 cell extract based on a 100 μg/mL extract load.
The concentrations of SET7 were measured in duplicate and interpolated from the SET7 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean SET7 concentration was determined to be 1,035 pg/mL.

Sandwich ELISA - Mouse SET7 ELISA Kit (AB279404)
Example of mouse SET7 standard curve in 1X Cell Extraction Buffer PTR.
The SET7 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

Product details
Mouse SET7 ELISA kit (ab279404) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Mouse SET7 protein in human cell and tissue extract samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse SET7 with 12 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
SET7, Histone-lysine N-methyltransferase SETD7, is a critical methyltransferase in epigenetic modification of histones. It specifically methylates lys4 of histone H3. It is homologous with many species throughout Mammalia, and as such is expected to cross-react with many species. The immunogen from which the antibodies in this kit were raised represents was a full-length recombinant protein with a minor N-terminal truncation.

Biological function summary
SET7 contributes to gene regulation by methylating histone H3 at lysine 4 (H3K4) a mark generally associated with transcriptional activation. It also methylates non-histone proteins such as p53 TAF10 and STAT3 affecting their activity and interactions. SET7 operates distinctively and does not need to be part of any complex to fulfill its function effectively in diverse cellular processes.
Pathways
SET7 modulates transcriptional pathways such as the p53 signaling pathway and the NF-kB pathway. It interacts with the p53 protein affecting cell cycle regulation and the response to DNA damage. The modification of NF-kB by SET7 can influence inflammation and immune responses. Through these pathways SET7 plays a role in maintaining cellular homeostasis and response to stress.
