Sandwich ELISA - Mouse SHP-2 ELISA Kit (PTPN11) (AB206309)
Interpolated concentrations of SHP-2 in PC12 (rat), L929 (mouse) and HeLa (human) cell extracts.
The concentrations of SHP-2 were measured in duplicate and interpolated from the SHP-2 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean SHP-2 concentration was determined to be 79.6 pg/μg PC12 cell extract, 37.3 pg/μg L929 cell extract, and 23 pg/μg HeLa cell extract.

Product details
Mouse SHP-2 ELISA Kit (PTPN11) (ab206309) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of SHP-2 (PTPN11) protein in cell culture extracts and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse SHP-2 (PTPN11) with 239 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
SHP-2 (Tyrosine-protein phosphatase non-receptor type 11 or PTPN11) is a widely expressed protein tyrosine phosphatase that functions in intracellular signal transduction. SHP-2 contains two phospho-tyrosine binding Src homology domains. SHP-2 is mutated in Noonan syndrome 1. Human, mouse and rat SHP-2 have >99% sequence identity on the amino acid level.

Biological function summary
The role of SHP2 extends to involvement in several signaling cascades such as the Ras/MAPK and PI3K/AKT pathways. It functions as an essential component within protein complexes that facilitate cell communication and response to external signals. The protein modulates growth factor signaling and cytokine signaling highlighting its significance in normal cell function and development. SHP2's statement in signaling processes makes it an important regulator of cellular dynamics.
Pathways
SHP2 participates in the Ras/MAPK and PI3K/AKT signaling pathways which are important for regulating cell growth survival and differentiation. Within these pathways SHP2 interacts with various signaling molecules including Grb2 Sos and Gab family adaptors. These interactions coordinate cellular responses to growth factors and other extracellular cues ensuring proper pathway activation and control. By serving as a critical mediator SHP2 integrates signals that are necessary for appropriate cellular outcomes.
