Product details

Abcam's Shh-N Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of mouse Shh-N in cell lysates and tissue lysates.

This assay employs an antibody specific for mouse Shh-N coated on a 96-well plate. Standards and samples are pipetted into the wells and Shh-N present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse Shh-N antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Shh-N bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Sonic Hedgehog N also known as Shh is a signaling molecule that plays an important role in cell communication during embryonic development. It is a protein with a mass of approximately 19 kDa. In mammals including mice Shh is mainly expressed in the developing central nervous system limbs and certain regions of the gastrointestinal tract. This molecule is locally secreted and operates by interacting with receptor proteins on the surface of target cells.
Biological function summary

Sonic Hedgehog N regulates the growth and patterning of tissues. It acts by binding to the Patched receptor part of the Hedgehog signaling complex which subsequently releases inhibition on another protein called Smoothened. This interaction facilitates the transcription of target genes involved in cell differentiation and tissue organization. Shh is also essential for the maintenance of stem cell populations in adult tissues.

Pathways

Sonic Hedgehog N is a central player in the Hedgehog signaling pathway which is both an important regulator of embryonic development and a fine tuner of adult homeostasis. The pathway interacts with proteins such as Gli which are transcription factors modulating gene expression downstream of Shh signaling. Shh also interfaces with the Wnt signaling pathway frequently contributing to coordinated control of cell proliferation and differentiation.

Sonic Hedgehog N has significant implications in cancers like basal cell carcinoma and medulloblastoma. Aberrant activation of the Shh pathway can lead to uncontrolled cell growth and tumor formation. Additionally defects in Shh signaling are associated with congenital disorders like holoprosencephaly where the structure of the brain is affected. Proteins like PTCH1 and SUFU which regulate or are regulated by Shh are also important to understanding its role in these diseases.

Sonic hedgehog protein. The C-terminal part of the sonic hedgehog protein precursor displays an autoproteolysis and a cholesterol transferase activity (PubMed : 7736596, PubMed : 7891723, PubMed : 8824192). Both activities result in the cleavage of the full-length protein into two parts (ShhN and ShhC) followed by the covalent attachment of a cholesterol moiety to the C-terminal of the newly generated ShhN (PubMed : 8824192). Both activities occur in the reticulum endoplasmic (PubMed : 21357747). Once cleaved, ShhC is degraded in the endoplasmic reticulum (PubMed : 21357747).. Sonic hedgehog protein N-product. The dually lipidated sonic hedgehog protein N-product (ShhNp) is a morphogen which is essential for a variety of patterning events during development. Induces ventral cell fate in the neural tube and somites (PubMed : 11430830, PubMed : 24863049). Involved in the patterning of the anterior-posterior axis of the developing limb bud (PubMed : 15315762). Essential for axon guidance (PubMed : 12679031). Binds to the patched (PTCH1) receptor, which functions in association with smoothened (SMO), to activate the transcription of target genes (By similarity). In the absence of SHH, PTCH1 represses the constitutive signaling activity of SMO (By similarity).
