Receptor for TNFSF13/APRIL and TNFSF13B/TALL1/BAFF/BLYS that binds both ligands with similar high affinity. Mediates calcineurin-dependent activation of NF-AT, as well as activation of NF-kappa-B and AP-1. Involved in the stimulation of B- and T-cell function and the regulation of humoral immunity (By similarity).

Mouse TACI ELISA Kit (TNFRSF13B) (ab206312) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of TACI (TNFRSF13B) protein in cell culture extracts, cell culture supernatant, plasma, serum, and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse TACI (TNFRSF13B) with 4 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (Pre-coated 384 well Microplate SimpleStep ELISA® ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Transmembrane activator and CAML interactor (TACI), also known as TNF receptor super family member 13B (TNFRSF13B), is a transmembrane receptor expressed primarily in mature B cells and is a regulator of B-cell homeostasis and immune response. Specifically, TACI binds both a proliferation-inducing ligand (APRIL) and B cell activating factor of the tumor necrosis factor family (BAFF) similarly with high affinity.Taci−/−mice have demonstrated that inadequate TACI signaling promotes the development of autoimmune diseases. This assay recognizesthe extracellular N-terminus of TACI.

Transmembrane Activator and CAML Interactor known as TACI also goes by the names TNFRSF13B and CD267. TACI is a receptor with a molecular weight of approximately 33 kDa. It is expressed on B cells a type of lymphocyte and to some extent on activated T cells. This receptor plays a part in the immune system helping manage the production and function of certain antibodies.
TACI involves the regulation of B cell function and survival. It binds to two ligands APRIL and BAFF and TACI is not part of any larger complex. Upon ligand binding TACI can promote antibody class-switching which adjusts the type of antibody that B cells produce. This action supports the immune response by allowing the body to adapt to different pathogens effectively.
Immune responses involve TACI in the BAFF-APRIL signaling pathway. The interplay between TACI and its ligands influences the NF-kB signaling cascade leading to B cell proliferation and survival. TACI interacts with other proteins such as BCMA and BAFF-R which are important in regulating B cell homeostasis and function ensuring balanced and effective immune responses.
Aberrant TACI function contributes to conditions like Common Variable Immunodeficiency (CVID) and Systemic Lupus Erythematosus (SLE). In CVID mutations in the TACI gene may lead to impaired antibody production resulting in frequent infections. In the context of SLE TACI's interactions with BAFF and APRIL can contribute to the overactivation of B cells causing autoimmune characteristics of the disease. TACI's role in these conditions makes it a focus for studying therapeutic interventions.

Interpolated concentrations of TACI in mouse serum, mouse plasma (citrate), and mouse plasma (EDTA).
The concentrations of TACI were measured in duplicate and interpolated from the TACI standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TACI concentration was determined to be 801.6 pg/mL in neat mouse serum,1744.4 pg/mL in neat mouse plasma (Citrate), 1766.4 pg/mL in neat mouse plasma (EDTA), and 16.2 ng/mL in neat A20 Cell Culture Supernatant.
Interpolated concentrations of TACI in mouse spleen and A20 cell extracts.
The concentrations of TACI were measured in duplicate and interpolated from the TACI standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TACI concentration was determined to be 5174.4 pg/mL in 4 mg/mL mouse spleen extract and 23 ng/mL in 150 μg/mL A20 cell extract.
