Sandwich ELISA - Mouse TECK ELISA Kit (CCL25) (AB269560)
Interpolated concentrations of native TECK in mouse thymus extract samples based on a 75 μg/mL extract load.
The concentrations of TECK were measured in duplicate and interpolated from the TECK standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TECK concentration was determined to be 658 pg/mL in thymus extract.

Sandwich ELISA - Mouse TECK ELISA Kit (CCL25) (AB269560)
Example of mouse TECK standard curve in 1X Cell Extraction Buffer PTR.
The TECK standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

Product details
Mouse TECK ELISA Kit (CCL25) (ab269560) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of TECK (CCL25) protein in tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse TECK (CCL25) with 7.4 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Mouse TECK (CCL25) is a CC chemokine produced by stromal cells in the thymus and epithelial cells of the small intestine. TECK is involved in human T-cell development inducing chemoattraction through CCR9 on immature pre-T cells and thymocytes. TECK induces the homing of several lymphocyte populations to the small intestine, including integrin a4b7+ gamma/delta T cells, Integrin aEb7+ CD8+ T cells, and IgA-producing plasma cells. In human cancer, TECK promotes the metastasis of melanoma cells, migration and invasion of some breast carcinomas, and mesenchymal stromal cells in multiple myelomas. TECK attracts CCR9+ monocytes and macrophages in rheumatoid arthritis and CCR9+ macrophages in atherosclerosis. Mouse TECK shares 50% ,81% and 61 % sequence homology with mouse, rat and cow, respectively

Biological function summary
This chemokine aids in the migration and homing of T-cell subsets particularly to the gut and small intestine hinting at its role in mucosal immunity. CCL25 does not belong to a larger multiprotein complex but it plays a standalone role in controlling immune surveillance and response. Its involvement contributes to the precise regulation of immune cell localization especially under inflammatory conditions.
Pathways
CCL25 participates significantly in the chemokine signaling pathway and the integrin-mediated adhesion pathway. It cooperates with proteins like CCR9 which is its receptor to activate downstream signaling cascades necessary for immune cell trafficking and adhesion. Through these pathways CCL25 ensures correct immune cell distribution throughout the body supporting effective immune surveillance and response.
