Sandwich ELISA - Mouse TFF3 ELISA Kit (AB253228)
Interpolated concentrations of native TFF3 in mouse plasma and cell samples.
The concentrations of TFF3 were measured in duplicates, interpolated from the TFF3 standard curves and corrected for sample dilution. Undiluted samples are as follows : plasma (citrate) 20%, plasma (EDTA) 20%, and plasma (heparin) 10%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TFF3 concentration was determined to be 1599.11 pg/mL plasma (citrate) in 2601.71 pg/mL in plasma (citrate), and 2822.63 pg/mL in plasma (heparin).

Mouse TFF3 ELISA Kit (ab253228) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of TFF3 protein in cit plasma, edta plasma, hep plasma, and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse TFF3 with 2.9 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Trefoil factor 3 is a protein that is encoded by the TFF3 gene. Members of the trefoil family can interact with mucins and have an influence on mucus viscosity. They also promote migration of epithelial cells, are linked to anti-apoptosis, induce cell scattering, trigger chemotaxis, and participate in immune responses. TFF3 is a marker of columnar epithelium and is expressed in a variety of tissues including goblet cells of the intestines and colon. It has been found that TFF3 is the most highly upregulated gene in 19 poorly differentiated endometrioid endometrial carcinomas (G3-EECs) compared with normal endometrium biopsies. Mouse TFF3 shares 85.2%, 94.8%, 82.5%, and 72.9% sequence homology with human, rat, bovine, and primate respectively.

Biological function summary
TFF3 plays an important role in protecting and repairing epithelial tissues particularly those lining the gastrointestinal tract. This protein promotes epithelial restitution by aiding cell migration and inhibits apoptosis contributing to tissue healing processes. While TFF3 does not form part of a large protein complex it often interacts with mucins specifically mucin 2 to bolster the protective mucus layer which is essential for shielding epithelial cells.
Pathways
Trefoil Factor 3 is notably involved in pathways associated with mucosal immune response and inflammation. A significant pathway involving TFF3 is the mucosal defense pathway where it works alongside other mucins and proteins like epidermal growth factor (EGF) to mediate regeneration and defense of epithelial tissues after injury. It also engages in the Wnt signaling pathway where it affects cell proliferation and migration critical for maintaining epithelial homeostasis.
