Interpolated concentrations of native TGF beta Receptor II in mouse serum, plasma and cell culture supernatant samples.

The concentrations of TGF beta Receptor II were measured in duplicates, interpolated from the TGF beta Receptor II standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma (citrate) 100%, plasma (EDTA) 100%, plasma (heparin) 50%, and lung supernatant 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TGF beta Receptor II concentration was determined to be 5.71 ng/mL in serum, 3.76 ng/mL in plasma (citrate), 4.36 ng/mL in plasma (EDTA), 6.30 ng/mL in plasma (heparin), and 2.14 ng/mL in and lung supernatant.

Interpolated concentrations of native TGF beta Receptor II in mouse lung extract based on a 2,000 μg/mL extract load and mouse spleen extract based on a 500 μg/mL extract load.

The concentrations of TGF beta Receptor II were measured in duplicate and interpolated from the TGF beta Receptor II standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TGF beta Receptor II concentration was determined to be 2.94 ng/mL in lung extract and 1.34 ng/mL in spleen extract.

Mouse TGF beta Receptor II ELISA kit (ab277719) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Mouse TGF beta Receptor II protein in mouse serum, plasma, cell and tissue culture supernatant, and cell and tissue culture extract samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse TGF beta Receptor II with 15.2 pg/mL sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Mouse TGF beta Receptor II is a transmembrane serine/threonine kinase that is formed with TGF-beta type I receptor, TGFBR1, the non-promiscuous receptor for the TGF-beta cytokines TGFB1, TGFB2 and TGFB3. TGF beta Receptor II transduces the TGFB1, TGFB2 and TGFB3 signal from surface of the cell to the cytoplasm. TGF beta Receptor II is involved in the regulation of cell cycle arrest in epithelial and hematopoietic cells, control of mesenchymal cell proliferation and differentiation, wound healing, extracellular matrix production, immunosuppression and carcinogenesis. Mouse TGF beta Receptor II shares 88.2%, 96.2%, 87.0%, and 91.7% sequence homology with human, rat, cow, and monkey, respectively.

Target data

Transmembrane serine/threonine kinase forming with the TGF-beta type I serine/threonine kinase receptor, TGFBR1, the non-promiscuous receptor for the TGF-beta cytokines TGFB1, TGFB2 and TGFB3. Transduces the TGFB1, TGFB2 and TGFB3 signal from the cell surface to the cytoplasm and is thus regulating a plethora of physiological and pathological processes including cell cycle arrest in epithelial and hematopoietic cells, control of mesenchymal cell proliferation and differentiation, wound healing, extracellular matrix production, immunosuppression and carcinogenesis. The formation of the receptor complex composed of 2 TGFBR1 and 2 TGFBR2 molecules symmetrically bound to the cytokine dimer results in the phosphorylation and the activation of TGFRB1 by the constitutively active TGFBR2. Activated TGFBR1 phosphorylates SMAD2 which dissociates from the receptor and interacts with SMAD4. The SMAD2-SMAD4 complex is subsequently translocated to the nucleus where it modulates the transcription of the TGF-beta-regulated genes. This constitutes the canonical SMAD-dependent TGF-beta signaling cascade. Also involved in non-canonical, SMAD-independent TGF-beta signaling pathways (By similarity).. Isoform RII-1.
