Metalloproteinase inhibitor that functions by forming one to one complexes with target metalloproteinases, such as collagenases, and irreversibly inactivates them by binding to their catalytic zinc cofactor. Acts on MMP1, MMP2, MMP3, MMP7, MMP8, MMP9, MMP10, MMP11, MMP12, MMP13 and MMP16. Does not act on MMP14 (By similarity). Also functions as a growth factor that regulates cell differentiation, migration and cell death and activates cellular signaling cascades via CD63 and ITGB1. Plays a role in integrin signaling.

Mouse TIMP1 ELISA Kit (ab196265) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of TIMP1 protein in cell culture supernatant, serum, plasma, and cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse TIMP1 with 6.2 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Tissue metalloproteinase inhibitor 1 (TIMP1or Metalloproteinase inhibitor 1) inhibits matrix metalloproteinases (MMP's) which cause degradation of the extracellular matrix. TIMP1 inhibits MMP's by forming a non-covalent bond complex at their catalytic zinc cofactor site and irreversibly inactivating them. TIMP1 also promotes cell proliferation and has some anti-apoptotic functions.

Tissue Inhibitor of Metalloproteinase 1 (TIMP1) is a protein with a molecular mass of approximately 28 kDa. The protein inhibits metalloproteinases by binding to them preventing the breakdown of the extracellular matrix. This action regulates extracellular environment and tissue remodeling. TIMP1 is also known by the name Erythroid-Potentiating Activity (EPA). It is expressed in various tissues including the liver and the brain and can be found in high levels in the blood serum.
TIMP1 plays several essential roles beyond inhibiting metalloproteinases. It supports cell proliferation and influences apoptosis. It is part of a larger TIMP family and does not function as part of a complex per se but interacts closely with matrix metalloproteinases (MMPs). By regulating MMP activity TIMP1 balances processes like tissue growth and inflammatory responses which are important for normal development and repair.
TIMP1 participates in the regulation of the matrix metalloproteinase pathway impacting tissue homeostasis and repair. It closely associates with proteins such as MMP-9 and MMP-2 within this pathway. TIMP1 also plays a role in cytokine signaling pathways which influence immune responses by interacting with other signaling molecules and receptors that govern these pathways.
TIMP1's regulation of MMPs links it to cancer and cardiovascular disease progression. Overexpression of TIMP1 associates with tumor growth and metastasis where it can modulate interactions with other proteins like MMP-9 leading to altered tissue invasion and angiogenesis. Additionally TIMP1 imbalance connects to tissue fibrosis in cardiovascular disorders working in tandem with proteins such as MMP-2 which affects the structural remodeling and function of tissues within the cardiovascular system.

Interpolated concentrations of TIMP1 in Mouse serum and plasma (citrate).
The concentrations of TIMP1 were measured in duplicate and interpolated from the TIMP1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean TIMP1 concentration was determined to be 4,495 pg/mL in Mouse serum and 4,210 pg/mL in Mouse plasma.
Titration of L929 Extract diluted 62.5 µg/mL to 4 µg/mL in Sample Diluent NS.

Comparison of secreted TIMP1 in unstimulated and PMA/PHA-stimulated L929 Cells.
L929 cells were grown in the absence (unstimulated) or presence of Phorbol Myristate Acetate (PMA) and phytohemagglutinin (PHA) (stimulated) for 3 days. TIMP1 was measured in 2-fold diluted cell culture supernatants of unstimulated and PMA/PHA stimulated L929 and cell culture media. Measured values were interpolated from the TIMP1 Standard Curve diluted in Sample Diluent NS and corrected for dilution factor. Mean of duplicate values +/-SD are graphed: 4.1 ng/mL unstimulated, 18.5 ng/mL stimulated. There was no detectable signal in media.
Interpolated concentration of native TIMP1 was measured in duplicate at different sample concentrations in 96-well vs. 384-well plates. Undiluted samples are 10% mouse serum. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.
